摘要
目的探讨miR-139-5p在肺腺癌细胞进展中的作用和机制。方法培养肺腺癌细胞A549、PC9、H292和正常人支气管上皮细胞BEAS-2B,采用qRT-PCR检测miR-139-5p。生物信息网站预测miR-139-5p的目标基因,分析找到与miR-139-5p相关性最高的基因CTNNB1。转染miR-139-5p inhibitor和oe-CTNNB1后,采用CCK-8实验、划痕实验、Transwell迁移实验和侵袭实验观察A549和PC9的增殖、迁移和侵袭能力,qRT-PCR和Western blot观察CTNNB1转录水平和蛋白质表达水平。结果与正常人支气管上皮细胞BEAS-2B相比,在肺腺癌细胞中miR-139-5p、CTNNB1高表达;敲低miR-139-5p后肺腺癌细胞的增殖、迁移及侵袭能力受抑制且CTNNB1转录水平及蛋白表达水平均下降,而过表达CTNNB1逆转了敲低miR-139-5p对肺腺癌细胞及CTNNB1表达的抑制作用。结论沉默miR-139-5p通过下调CTNNB1抑制肺腺癌细胞的增殖、迁移及侵袭能力。
Objective To explore and analyze the role and mechanism of miR-139-5p in the progression of lung adenocarcinoma cells.Methods Lung adenocarcinoma cells were cultured A549,PC9 and H292 and normal human bronchial epithelial cells BEAS-2B,and miR-139-5p was detected by qRT-PCR.miR-139-5p target genes were predicted by Bioinformatics website,and analyzed to find CTNNB1,the gene with the highest correlation with miR-139-5p.miR-139-5p inhibitor and oe-CTNNB1 were transfected and then analyzed by CCK-8 assay,scratch assay.Transwell migration assay and invasion assay were used to observe the proliferation,migration and invasion ability of A549 and PC9.qRT-PCR and Western blot were used to observe the transcription level and protein expression level of CTNNB1.Results Compared with the control group,miR-139-5p and CTNNB1 were highly expressed in lung adenocarcinoma cells.The proliferation,migration and invasion of lung adenocarcinoma cells were inhibited by knockdown of miR-139-5p,and the transcription and protein expression of CTNNB1 were decreased.The overexpression of CTNNB1 reversed the inhibitory effect of knockdown of miR-139-5p on the expression of lung adenocarcinoma cells and CTNNB1.expression.Conclusion miR-139-5p is highly expressed in lung adenocarcinoma and targets and promotes CTNNB1 to enhance the proliferation,migration and invasion of lung adenocarcinoma cells.
