大豆苷元对肥胖小鼠脂质代谢和肠道菌群的调节作用研究

摘要 为阐明大豆苷元(daidzein,DAI)对肥胖小鼠脂质代谢和肠道菌的调节作用,将18只小鼠随机分为3组,每组6只,分别为模型组(DIO)、DAI组(DAI)和空白对照组(CON)。建立小鼠肥胖模型,其中DAI组灌胃给予DAI,剂量为100 mg/kg,DIO和CON组给予等体积的0.5%CMC-Na溶液。试验期间每周记录小鼠空腹体重和累积采食量,试验结束后,检测血清中甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)和低密度脂蛋白胆固醇(LDL-C)的含量,称量各组小鼠的体重变化、观察附睾脂肪组织变化,利用高通量测序分析小鼠肠道微生物的多样性、物种组成和菌群差异。结果显示,DIO组和DAI组的平均体重分别是CON组的151.82%和151.46%,表明肥胖模型建立成功。与DIO组相比,DAI组小鼠的体重显著降低(P<0.01),TG、TC、LDL-C的含量显著降低(P<0.01),而HDL-C的含量显著升高(P<0.01);与DIO组相比,DAI组eWAT中的脂肪细胞大小均匀,形态正常。与DIO组相比,DAI组小鼠肠道中乳杆菌属(Lactobacillus)、异杆菌属(Allobaculum)的丰度升高,而脱硫弧菌属(Desulfovibrio)、穆齐螺旋菌(Mucispirillum)的丰度降低。随机森林的结果显示,醋酸杆菌属(Acetobacter)、嗜黏蛋白阿克曼菌(Akkermansia)和[普雷沃氏菌属Prevotella]可能是DAI调节小鼠脂质代谢的关键肠道菌。研究结果表明DAI可以抑制肥胖小鼠的体重增加,改善肥胖小鼠的脂质代谢和菌群结构。 To elucidate the regulating effect of daidzein(DAI)on lipid metabolism and gut microbiota in obese mice,18 mice were randomly divided into model group(DIO),DAI group(DAI)and control group(CON).Obesity model was established,in which the DAI group was given DAI by gavage at a dose of 100 mg/kg,DIO group and CON group were given equal volumes of 0.5%CMC-Na solution.After the experiment,the contents of TG,TC,HDL-C and LDL-C in sera were detected,the body weight and fat weight of mice in each group were weighed.The changes of epididymal adipose tissue were observed by HE staining and the diversity,composition and difference of gut microbiota were analyzed by high-throughput sequencing.The results showed that the average weight of DIO group and DAI group were 151.82%and 151.46%compared to CON group,respectively.Therefore,the obesity model was successfully established.Compared with DIO group,the body weight,TG,TC and LDL-C of the DAI group was significantly reduced(P<0.01)and the content of HDL-C was significantly increased(P<0.01).Compared with the DIO group,the adipocytes were uniform size and normal morphology in DAI group.Compared with the DIO group,the abundances of Lactobacillus and Allobaculum were increased,while the abundances of Desulfovibrio and Mucispirillum were decreased in DAI group.The results of the random forest showed that Acetobacter,Akkermansia and Prevotella were regarded as the key bacteria that regulates lipid metabolism in mice.In conclusion,DAI can suppress the weight gain and improve lipid metabolism and gut microbiota structure in obese mice.
