摘要
三角槭叶色美丽,用途广泛,观赏价值和经济价值极高。为快速获得大量优质种苗,实现工厂化育苗,本研究以三角槭带芽茎段为外植体,对外植体材料的最佳采集时间、消毒方式、茎段腋芽诱导和生根诱导培养基配方进行筛选,建立了三角槭一步成苗体系。为进一步探究三角槭幼苗的生根机制,对不定芽的根原基进行了细胞学分析和生根发育时期关键基因相对表达量鉴定。结果表明,6月采集的外植体使用75%乙醇消毒20 s和0.1%氯化汞消毒15 min的组合灭菌方式效果最佳。茎段腋芽诱导最适培养基为MS培养基+0.5 mg/L吲哚丁酸(IBA),诱导率达82.3%。不定芽生根诱导最适培养基为1/2 MS+0.6 mg/L IBA,生根率达88.7%,平均根数达4.9条。三角槭不定根原基的发生与IBA密切相关,在0.6 mg/L IBA的作用下,根原基发生明显,生根发育关键基因AbPIN1、AbPIN2和AbPIN3的相对表达量显著升高。本研究为三角槭新品种规模化繁育提供了技术参考,为探究三角槭生根机制奠定了基础。
The Acer buergerianum,renowned for its ornamental foliage and extensive applications,possesses significant aesthetic and economic value.To facilitate the rapid production of high-quality seedlings and achieve industrial-scale propagation,this study employed stem segments with buds as explants to screen for optimal collection times,sterilization methods,and medium formulations for axillary bud induction and root induction.A comprehensive tissue culture rapid propagation system for Acer buergerianum was established.Further investigation into the root formation mechanism involved cytological analysis of adventitious root primordia and the expression levels of key genes during root development.The findings indicated that a combination of 75% ethanol disinfection for 20 seconds followed by 0.1%mercuric chloride disinfection for 15 minutes was the most effective sterilization method for explants collected in June.The optimal medium for axillary bud induction was MS+0.5 mg/L indole-3-butyric acid(IBA),achieving an induction rate of 82.3%.For adventitious root induction,the optimal medium was 1/2 MS+0.6 mg/L IBA,resulting in a rooting rate of 88.7% with an average of 4.9 roots per plant.The initiation and growth of adventitious root primordia in Acer buergerianum were closely associated with auxin IBA.At a concentration of 0.6 mg/L IBA,the formation of root primordia was significantly enhanced,and the relative expression levels of key genes AbPIN1,AbPIN2,and AbPIN3 during root development increased markedly.This study provides a technical reference for large-scale propagation of new varieties of Acer buergerianum and establishes a foundation for exploring its root formation mechanism.
