大黄粉对脓毒症小鼠肠道炎性因子水平的影响

摘要 目的:探究大黄粉对脓毒症小鼠的疗效。方法:60只雄性ICR小鼠,随机选取40只,采用盲肠结扎穿孔法(CLP)制备脓毒症模型后再随机分为大黄粉组(采用大黄粉灌胃处理)和模型组(灌胃等量生理盐水),每组各20只。余20只作为对照组(只开腹,不作盲肠结扎穿孔处理)。采用ELISA检测小鼠血清和肠组织中IL-6、IL-10、IL-1β和TNF-α水平;实时荧光定量PCR(RT-PCR)检测小鼠肠组织中紧密连接相关基因(Claudin-4、ZO-1和Occludin)mRNA水平;Western blot实验检测小鼠肠组织中炎症小体相关蛋白(ASC和NLRP3)表达水平。结果:与对照组相比,模型组血清和肠组织中IL-6和IL-10水平下降,IL-1β和TNF-α水平升高(P<0.05);Claudin-4、ZO-1、Occludin mRNA水平降低(P<0.05);ASC和NLRP3蛋白表达水平升高(P<0.05)。与模型组相比,大黄粉组血清和肠组织中IL-6和IL-10水平升高,IL-1β和TNF-α水平降低(P<0.05);Claudin-4、ZO-1、Occludin mRNA水平升高(P<0.05);ASC和NLRP3蛋白表达水平降低(P<0.05)。结论:大黄粉通过减轻脓毒症小鼠肠组织中炎性因子表达和炎症小体的激活,从而恢复肠道屏障系统功能,抑制脓毒症进展。 Objective:To investigate the therapeutic effect of rhubarb powder on sepsis mice.Method:Sixty male ICR mice were selected.Forty mice were randomly chosen to establish sepsis models using the cecal ligation and puncture(CLP)method,and then randomly divided into the rhubarb powder group(treated with rhubarb powder by gavage)and the model group(gavaged with an equal volume of normal saline),with 20 mice in each group.The remaining 20 mice served as the control group(undergoing sham operation,i.e.,laparotomy followed by immediate abdominal closure without cecal ligation and puncture).ELISA was used to detect the levels of IL-6,IL-10,IL-1β,and TNF-α in mouse serum and intestinal tissues;real-time fluorescent quantitative PCR(RT-PCR)was employed to determine the mRNA levels of tight junction-related genes(Claudin-4,ZO-1,and Occludin)in mouse intestinal tissues;Western blot was applied to measure the expression levels of inflammasome-related proteins(ASC and NLRP3)in mouse intestinal tissues.Results:Compared with the control group,the model group showed decreased levels of IL-6 and IL-10,and increased levels of IL-1β and TNF-α in serum and intestinal tissues(P<0.05);the mRNA levels of Claudin-4,ZO-1,and Occludin were reduced(P<0.05);the protein expression levels of ASC and NLRP3 were elevated(P<0.05).Compared with the model group,the rhubarb powder group exhibited increased levels of IL-6 and IL-10,and decreased levels of IL-1β and TNF-α in serum and intestinal tissues(P<0.05);the mRNA levels of Claudin-4,ZO-1,and Occludin were increased(P<0.05);the protein expression levels of ASC and NLRP3 were decreased(P<0.05).Conclusion:Rhubarb powder can restore the function of the intestinal barrier system and inhibit the progression of sepsis by reducing the expression of inflammatory factors and the activation of inflammasomes in the intestinal tissues of sepsis mice.
