摘要
利用双歧杆菌对海藻进行发酵并以单因素实验考察其高密度发酵条件,优选出发酵条件为:石莼粉当做发酵藻体、石莼粉添加量2%、发酵氮源为1%大豆蛋白胨、发酵时间为26 h,发酵温度37℃。石莼粉发酵液分别进行了鸡胚绒毛尿囊膜实验及细胞急性毒性测试,考察其刺激性、安全性。进行了细胞紧致抗皱实验、舒缓实验、美白实验、抗氧化实验,考察其功效。结果显示,所得的石莼粉发酵液无刺激性,细胞无毒性浓度高,浓度为5%的石莼粉发酵液可促使Ⅰ型胶原蛋白上调32.58%±1.33%,10%的石莼粉发酵液对NO及黑色素的相对抑制率分别高达67.64%±3.99%和55.76%±0.11%,6.25%的石莼粉发酵液对ABTS自由基的清除率可达58.29%±4.92%。结果表明,石莼粉发酵液是一种高安全性、有优异抗皱、舒缓、美白功效及体外抗氧化能力的发酵产物,实现了高值化开发,有极大潜力应用于化妆品、食品等行业。
Bifidobacteria were used to ferment the seaweed and its high-density fermentation conditions were investigated by single factor experiments.The optimal fermentation conditions were determined as follows:Ulva lactuca powder as the fermenting algal body,Ulva lactuca powder addition amount of 2%,fermentation nitrogen source of 1%soy peptone,fermentation time of 26 h,and fermentation temperature of 37℃.The fermentation broth of Ulva lactuca powder was subjected to chicken embryo chorioallantoic membrane experiments and acute cytotoxicity tests to investigate its irritancy and safety.The cell tightening and anti wrinkle experiments,soothing experiments,whitening experiments,and antioxidant experiments were conducted,and their efficacy were investigated.The results showed that the fermentation broth of Ulva lactuca powder obtained was non irritating and had a high concentration of non-toxic cells.A 5%concentration of fermentation broth of Ulva lactuca powder could promote an upregulation of type Ⅰ collagen protein by 32.58%±1.33%.The relative inhibition rates of NO and melanin in 10% fermentation broth of Ulva lactuca powder were as high as 67.64%±3.99% and 55.76%±0.11%,respectively.The clearance rate of ABTS free radicals in 6.25% fermentation broth of Ulva lactuca powder could reach 58.29%±4.92%.The results indicate that the fermentation broth of Ulva lactuca powder is a highly safe and effective fermentation product with excellent anti wrinkle,soothing,whitening effects,and in vitro antioxidant capacity.It has achieved high-value development and has great potential for the application in industries such as cosmetics and food.
