激动药的拮抗参数pA_(2)计算模型研究

摘要 目的 探索医学机能实验课“药物量效曲线绘制及竞争性拮抗药pA_(2)的测定”中拮抗参数pA_(2)值的计算模型。方法 以牛蛙腹直肌为标本,乙酰胆碱(ACh)为激动药,筒箭毒碱为竞争性拮抗药,观察随着ACh剂量的增加,肌肉收缩效应的变化。应用Excel软件创建pA_(2)值计算模型,随机收集25组学生实验数据,t检验分析传统坐标法与模型处理所得的pA_(2)值。结果 其中14组数据第1条量效曲线最大效应较大数据的坐标法估算值与模型计算值分别为(6.39±0.35)和(6.37±0.34);而余下10组数据的分别为(6.15±0.50)和(6.42±0.57)。t检验分析结果显示两种方法计算所得pA_(2)值无显著性差异(P>0.05)。结论 本模型可用于高校医学机能实验课中随堂绘制药物量效曲线并计算pA_(2)值,有效提升数据处理的及时性、规范性和准确度,便于老师实时分析实验结果,提高实验效率和教学质量。 Objective To explore the calculation model of the antagonistic parameter pA,in the medical functional experi-ment course“determination of dose-effect relationship and pA_(2) of drugs”.Methods The ACh effect on the rectus abdominis muscle of bullfrogs was observed,while tubocurarine was used as a competitive antagonist.An excel calculation model of pA_(2) was created.The t-test was used to analyze the pA_(2) data from 25 random experimental group,in which traditional coordinate method or model treatment method were conducted.Results The estimated values of the coordinate method and the calculated values of the model for pA_(2) with the largest maximum effect of the first dose-effect curve(6.39±0.35)and(6.37±0.34)from 14 groups;And pA,from other 10 groups were(6.15±0.50)and(6.42±0.57),respectively.The results of t-test analysis showed that there was no significant difference in the pA_(2) values calculated by the two methods(P>0.05).Conclusion This model can be used for calculating pA,in medical functional experimental courses to effectively improve the timeliness,standardi-zation,and accuracy of data processing,then lecturers can analyze the experimental results in real-time to improve experimental efficiency and teaching quality.
