摘要
目的 探究新型近红外荧光探针ESS65-Br在胃部的成像特性,开发具有特异性的胃部成像荧光探针,为外科术中导航提供参考。方法 选择雄性CD-1小鼠3只,鼠龄4周;体质量25~30 g。借助海洋光学测量系统对新型近红外荧光探针ESS65-Br的光密度与荧光强度进行测定,进而剖析其光学特性。将新型近红外荧光探针ESS65-Br注射至CD-1小鼠体内,小动物活体成像动态检测小鼠全身显影情况,在注射后4 h,对小鼠体内的腹腔、胃及取出的腹部脏器进行了近红外荧光成像的观察,并测量了离体脏器的信号与背景比值(SBR)。结果 新型近红外荧光探针ESS65-Br的最大吸收峰值处于641 nm,荧光峰值则在700 nm。在小鼠体内,该探针于胃部呈现出明显的荧光信号,而心、肺、肝、胰、脾、肾、十二指肠及小肠等器官均未检测到荧光信号。对离体器官进行SBR分析结果表明,胃的荧光强度值为6.46±0.84,胃的SBR为6.33,胃部SBR显著高于其他器官。结论 新型近红外荧光分子探针ESS65-Br在胃部展现出优异的特异性与靶向性,其定位精准且成像清晰,有望为外科医师术中实时精准切除胃肿瘤提供一定的借鉴。
Objective To explore the imaging characteristics of novel near-infrared fluorescent probe ESS65-Br in stomach,and develop a specific gastric imaging fluorescent probe for providing reference in surgical navigation.Methods Three male CD-1 mice(aged 4 weeks with body mass of 25-30 g)were sacrificed.The optical density and fluorescence intensity of novel near-infrared fluorescent probe ESS65-Br were measured by marine optical measurement system,and its optical properties were analyzed.The novel near-infrared fluorescent probe ESS65-Br was injected into CD-1 mice,and whole body images of the mice were dynamically detected by small animal in vivo imaging.At 4th hour after injection,the abdominal cavity and stomach in vivo and abdominal organs removed in vitro were observed by near-infrared fluorescence imaging,and the signal-to-background ratio(SBR)of isolated organs was measured.Results The maximum absorption peak of novel near-infrared fluorescent probe ESS65-Br was at 641 nm,and the fluorescence peak was at 700 nm.In vivo,the probe showed obvious fluorescence signal in stomach,while no fluorescent signals were detected in organs such as the heart,lung,liver,pancreas,spleen,kidney,duodenum and small intestine.The results of SBR analysis of isolated organs showed that the fluorescence intensity of the stomach was 6.46±0.84,and the SBR of the stomach was 6.33.The SBR of the stomach was significantly higher than that of other organs.Conclusion It is demonstrated that the novel near-infrared fluorescent probe ESS65-Br showed specificity and targeting in stomach,with precise positioning and clear imaging.It is expected to provide a reference for surgeon to accurately remove gastric tumor in real time during surgery.
