北刘寄奴外敷治疗不完全骨折的网络药理学研究

摘要 目的基于网络药理学探讨北刘寄奴外敷治疗不完全骨折的分子机制。方法根据类药性(DL)在中药系统药理学数据库与分析平台(TCMSP)数据库中筛选北刘寄奴的活性成分,在ChemSpider数据库中获得这些成分的2D结构后导入SwissTargetPrediction平台以预测其作用靶点,随后在GeneCards数据库中获得不完全骨折靶点,两者靶点做交集处理,并对交集靶点进行蛋白质-蛋白质相互作用(PPI)分析、GO功能富集分析与KEGG通路富集分析。结果共得到北刘寄奴治疗不完全骨折的交集靶点218个。GO功能富集分析显示北刘寄奴治疗不完全骨折涉及对外源刺激的反应等生物学过程;KEGG通路富集分析显示北刘寄奴可能通过PI3K-Akt等信号通路治疗不完全骨折。结论北刘寄奴通过PI3K-Akt等信号通路促进不完全骨折的愈合。 Objective To investigate the molecular mechanism of Siphonostegiae herba external application in the treatment of incomplete fracture based on network pharmacology.Methods According to the drug-like properties(DL),the active components of Siphonostegiae herba were screened in the traditional Chinese medicine systems pharmacology database and analysis platform(TCMSP)database,and the 2D structures of these components were obtained in the ChemSpider database,and imported into the SwissTargetPrediction platform to predict their targets.Subsequently,the incomplete fracture targets were obtained in the GeneCards database,and the two targets were intersected.Protein-protein interaction(PPI)analysis,GO function enrichment analysis and KEGG pathway enrichment analysis were performed on the intersection targets.Results A total of 218 intersection targets of Siphonostegiae herba in the treatment of incomplete fractures were obtained.GO function enrichment analysis showed that the treatment of incomplete fracture by Siphonostegiae herba involved biological processes such as response to exogenous stimulation;KEGG pathway enrichment analysis showed that Siphonostegiae herba may treat incomplete fracture through PI3K-Akt and other signaling pathways.Conclusion Siphonostegiae herba promotes the healing of incomplete fracture through PI3K-Akt signaling pathway.
