摘要
目的通过对补中益气汤联合顺铂(cisplatin,DDP)对非小细胞肺癌荷瘤裸鼠移植瘤Wnt/JNK信号通路影响的研究,探讨补中益气汤增强顺铂敏感性的分子机制。方法培养A549细胞,将其接种于裸鼠腋窝皮下并将所有裸鼠随机等分为5组,分别是A549对照荷瘤组(a)、A549荷瘤+DDP组(b)、A549荷瘤+DDP+低剂量中药组(c)、A549荷瘤+DDP+中剂量中药组(d)、A549荷瘤+DDP+高剂量中药组(e)。测量并记录每组移植瘤的体积,测量每组移植瘤重量并计算抑瘤率。应用蛋白印迹法检测移植瘤组织中Wnt11、Rac1、JNK、p-JNK、c-Jun、p-c-Jun、P-gp蛋白的表达。结果补中益气汤可同时缩小A549细胞株荷瘤裸鼠移植体积,减轻移植瘤重量,提高顺铂抑瘤率,变化呈现显著的时间依赖性和剂量依赖性。与DDP组相比,DDP联合低、中、高剂量中药组移植瘤的Wnt11、Rac1、p-JNK、p-c-Jun、P-gp蛋白表达均下调(P<0.05),JNK、c-Jun蛋白表达无明显变化(P>0.05)。结论补中益气汤可抑制A549细胞株荷瘤裸鼠移植瘤生长,同时下调移植瘤Wnt11、Rac1、p-JNK、p-c-Jun、P-gp蛋白的表达,推测其可通过抑制Wnt/JNK信号通路来增强顺铂的敏感性。
Objective To explore the molecular mechanism of Buzhong Yiqi Decoction(补中益气汤)enhancing cisplatin sensitivity,by researching the effect of Buzhong Yiqi Decoction combined with cisplatin on the Wnt/JNK signaling pathway of non-small cell lung cancer transplanted tumors in nude mice.Methods A549 cells were cultured,inoculated in the axillary subcutis of nude mice and randomly divided into 5 groups:A549 control tumor group(a),A549 tumor+cisplatin group(b),A549 tumor+cisplatin+low-dose traditional Chinese medicine(c),A549 tumor+cisplatin+medium-dose traditional Chinese medicine(d),and A549 tumor+cisplatin+high dose traditional Chinese medicine(e).The volume of transplanted tumors in each group were measured and recorded,and the weights of transplanted tumors in each group were measured and the tumor inhibition rates were calculated.Western blotting was applied to detect the expressions of Wnt11,Rac1,JNK,p-JNK,c-Jun,p-c-Jun and P-gp proteins in transplanted tumor tissues.Results Buzhong Yiqi Decoction could simultaneously reduce the graft volume of A549 cell line hormonal nude mice,reduce the weight of graft tumor,and increase the tumor suppression rate of cisplatin,and the changes showed significant time-dependent and dose-dependent.Compared with those of the cisplatin group,the expressions of Wnt11,Rac1,p-JNK,p-c-Jun and P-gp proteins were down-regulated in traditional Chinese medicine low,medium and high dose groups(P<0.05),however,there was no significant change in the protein expressions of JNK or c-Jun(P>0.05).Conclusion Buzhong Yiqi Decoctionto can simultaneously down-regulate the expressions of Wnt11,Rac1,p-JNK,p-c-Jun and P-gp proteins,it is speculated whether increased sensitivity of cisplatin is induced by inhibiting the Wnt/JNK signaling pathway.
