流式细胞仪数据采集系统验证测试平台设计与实现 被引量:1

Design and Implementation of Verification and Testing Platform for Flow Cytometer Data Acquisition Systems

摘要 针对现有流式细胞仪数据采集系统性能测试手段匮乏、易受干扰等问题,研制了一种基于单片机的验证测试平台。该平台基于流式细胞仪检测信号特征,通过建立查找表并采用内存直访数模转换方法生成高斯包络的模拟细胞信号。利用单片机生成随机数以控制信号的随机产生,模拟了流式细胞仪检测事件发生的泊松过程。生成的信号再经匹配电路形成适用于现有流式细胞仪数据采集系统的目标信号。实验结果表明,该验证测试平台可以模拟白细胞通过检测光斑时产生的高斯包络信号及检测事件间的泊松过程,并由数据采集系统提供的增益信号动态调整平台输出信号的幅值。该验证测试平台实现了流式细胞仪事件信号的精确模拟,显著提高了数据采集系统性能验证的准确性,同时有效避免了液流、光学等干扰因素对数据采集系统性能评估的影响。 A microcontroller-based verification and testing platform was developed to address the problems of lack of performance testing methods and the influence of multiple interference parameters in the existing flow cytometer data acquisition system testing and validation platforms.The platform establishes a lookup table based on the characteristics of flow cytometer detecting signals and uses memory direct access and digital to analog conversion to generate simulated cell signals featuring Gaussian envelopes.It uses a microcontroller to generate random numbers to control the random generation of signals to simulate the Poisson process of flow cytometry detecting events.By use of matching circuits,it generates target signals for flow cytometer data acquisition systems.The experimental results show that the developed validation and testing platform can simulate the Gaussian shaped signal generated by white blood cells passing the laser spot and the Poisson process between detecting events,and dynamically adjust the amplitude of the platform output signal through the gain signal provided by the data acquisition system.By simulating the events signals,this platform improves the accuracy of functional performance verification of the data acquisition system and effectively avoids the influence of other interference factors such as fluid flow and optics on the performance evaluation of the data acquisition system.
