摘要
多杀性巴氏杆菌(Pasteurella multocida,Pm)是一种常见致病菌,可以引起多种禽类、家畜、野生动物,甚至人类的感染。本实验室先前从患病海南黑山羊组织中分离出1株D型Pm。为进一步了解该菌株的基因组特征,明确其毒力相关基因和遗传进化关系。为Pm感染疾病的预防和临床治疗提供参考。将该菌株命名为“Pm HN01”株,使用SMRT技术和Illumina测序技术相结合的方法进行全基因组测序。使用生物信息学、PCR鉴定和系统进化树等方法对Pm HN01株的基因组特征、毒力相关基因和遗传进化关系进行分析。将测序后的基因组信息提交至NCBI,获得登录号CP037861。Pm HN01株与其他巴氏杆菌具有相似的基因组谱,大小为2349898 bp,GC含量为40.25%,预测到2401个编码基因。在GO、KEGG和COG分析中分别有1651、2227和1912个基因注释。在PHI、VFDB和T3SS分析中,分别有121、43和84个基因注释。此外,选取23个毒力基因引物对其进行PCR鉴定,结果发现Pm HN01株含有exbB、exbD、tbpA、toxA、fimA等12个毒力基因,同北京和新疆等国内菌株的进化关系最近,同美国、瑞士、丹麦等国外菌株的进化关系最远。本研究首次获得了山羊源D型Pm HN01株的完整基因组信息,为Pm的基因组数据库增添了材料,丰富了Pm的宿主类型。进一步完善了Pm HN01株的毒力基因和系统进化信息,为后续Pm的流行区域和致病性研究提供了理论参考。
Pasteurella multocida(Pm)is a common pathogen,which causes infection in many animals including poultry,livestock,wild lives and even human beings.The capsular serotype D Pm strain HN01 was isolated from Hainan black goat in our previous study.The objectives of the present study were to investigate its genomic characteristics,virulence-associated genes and genetic evolution for further providing a reference for prevention and clinical treatment of pasteurellosis.The whole genome of the Pm strain HN01 was sequenced by the combination of SMRT technology and Illumina sequencing technology.The genomic information was then submitted to NCBI with the accession number CP037861.The results showed that the Pm strain HN01 had a genomic profile similar to other Pasteurella strains.The genome size of the Pm strain HN01 was 2349898 bp and the GC content was 40.25%.A total of 2401 encoding genes were predicted via bioinformatics.There were 1651,2227 and 1912 gene annotations in GO,KEGG and COG analysis,respectively.In PHI,VFDB,and T3SS analysis,there were 121,43 and 84 gene annotations,respectively.In addition,the Pm strain HN01 contained 12 virulence genes such as exbB,exbD,tbpA,toxA and fimA,etc.The phylogenetic analysis revealed that the Pm strain HN01 had the closest evolutionary relationship with domestic strains isolated from Beijing and Xinjiang and the farthest evolutionary relationship with foreign strains such as the United States,Switzerland and Denmark.In summary,the complete genome information of the goat-derived capsular serotype D Pm strain HN01 was obtained for the first time,which added materials to the genome database and enriched the host types of Pm.The completion of the virulence-associated genes and phylogenetic information of the Pm strain HN01 provided a reference for the future study of the epidemic area and pathogenicity of Pm.
