摘要
目的:探讨内源性硫化氢(H2S)是否参与调控肝硬化大鼠门静脉压力,以及与一氧化氮(NO)/一氧化氮合酶(NOS)体系和一氧化碳(CO)/血红素加氧酶-1(HO-1)体系之间相互作用。方法:将Sprague-Dawley大鼠随机划分成四组,分别为空白对照组(N组),肝硬化模型组(HC组)、肝硬化+硫氢化钠组(HC+NaHS组)和肝硬化+炔丙基甘氨酸组(HC+PPG组)。分别采用敏感硫电极法、硝酸还原酶法、联二亚硫酸盐还原法测定血浆中H2S、NO和CO含量,以门静脉插管法测定四组大鼠门静脉压力,Western blot测定肝组织中内皮型一氧化氮合酶(eNOS)、血红素加氧酶-1(HO-1)及胱硫醚-γ-裂解酶(CSE)蛋白的表达水平。结果:与N组比较,HC组血浆中H2S的水平及肝组织中CSE蛋白水平均显著下降,门静脉压力及NO/CO浓度、HO-1蛋白表达显著上升,e NOS蛋白显著下降。与HC组对比,当外源性给予NaHS后,血浆中H2S以及肝组织中CSE蛋白表达水平显著升高(P<0.01),大鼠门静脉压力、血浆中NO、CO的浓度及肝组织中eNOS和HO-1蛋白表达水平显著降低;当外源性给予H2S抑制剂PPG后,血浆中H2S的水平及肝组织中CSE蛋白表达水平显著降低,但门静脉压力却显著上升,血浆中NO/CO体系表达显著上升。结论:内源性H2S在肝硬化门静脉压力的调节中发挥保护作用,而H2S/CSE体系、NO/NOS体系和CO/HO-1体系则是通过网状关系协同参与门静脉高压的发生发展。
Objective Investigation into the Role of Endogenous Hydrogen Sulfide(H2S)in Regulating Portal Pressure in a Rat Model of Liver Cirrhosis and Its Interplay with the Nitric Oxide(NO)/Nitric Oxide Synthase(NOS)System and the Carbon Monoxide(CO)/Heme Oxygenase-1(HO-1)System.Methods Sprague-Dawley rats were randomly split into four groups:blank control group(N),hepatic cirrhosis model group(HC),HC+NaHS,HC+PPG.The contents of H2S,NO,and CO in plasma were measured by sensitive sulfur electrode,nitrate reductase,and bisulfite reduction,portal vein pressure was measured in four groups of rats by portal cannulation,and protein expression levels of eNOS,HO-1,and cystathionineγ-lyase(CSE)in liver tissue by Western blot.Results Compared with the N group,the levels of H2S in plasma and CSE protein in liver tissue of the HC group were significantly decreased,while the portal vein pressure and the concentrations of NO/CO,as well as the expression of HO-1 protein,were significantly increased,and the expression of eNOS protein was significantly decreased.Compared with the HC group,after exogenous administration of NaHS,the levels of H2S in plasma and the expression of CSE protein in liver tissue were significantly increased(P<0.01),and the portal vein pressure,concentrations of NO and CO in plasma,and the expression levels of eNOS and HO-1 protein in liver tissue were significantly decreased;after exogenous administration of the H2S inhibitor PPG,the levels of H2S in plasma and the expression of CSE protein in liver tissue were significantly decreased,but the portal vein pressure was significantly increased,and the expression of NO/CO synthase system was significantly increased.Conclusion Endogenous H2S plays an important protective role in the regulation of portal pressure in cirrhosis,while H2S/CSE,NO/NOS and CO/HO-1 are involved in the treatment of portal hypertension through reticulation.
