碳源种类及浓度对杜氏盐藻生长和代谢产物的影响研究

摘要 杜氏盐藻中含有丰富的营养物质,包括具有极高经济和医用价值的β胡萝卜素、作为生物燃料的油脂等。但是杜氏盐藻的产量和生产力较低,对其商业化生产有一定的限制。通过添加3种不同碳源(葡萄糖、乙酸钠、甘油),并设置不同浓度,研究不同碳源条件对杜氏盐藻的生长情况及代谢产物的积累情况,以期寻找较好的杜氏盐藻生长和代谢产物积累条件。结果显示:100 mmol·L^(-1)乙酸钠兼养培养下对杜氏盐藻的促生长效果最为明显,生物量可提高59%;150 mmol·L^(-1)葡萄糖兼养培养下对杜氏盐藻的促代谢产物积累效果最为明显,β胡萝卜素产量可提高246%,油脂产量提高63.3%,可溶性蛋白含量提高61.8%,可溶糖含量提高94.4%。本研究可以一定程度解决杜氏盐藻生长缓慢、代谢产物合成量少的问题,为获取高产β胡萝卜素的海洋藻株培养提供借鉴。 Dunaliella salina is rich in nutrients,including β-carotene with high economic and medical value,lipids as biofuel feedstock and so on.However,the yield and productivity of D.salina are relatively low,which limits its commercial application.In this study,three different carbon sources(glucose,sodium acetate and glycerol)with different concentrations were added to study the growth of D.salina and the accumulation of metabolites under different carbon sources conditions,in order to find better conditions for the growth of D.salina and the accumulation of metabolites.The results showed that the growth promotion effect of 100 mmol·L^(-1) sodium acetate culture was the most obvious,and the biomass could be increased by 59%.The promotion of metabolite accumulation in D.salina with 150 mmol·L^(-1) glucose was most obvious,which could increase β-carotene production by 246%,lipid content by 63.3%,soluble protein content by 61.8%,and soluble sugar content by 94.4%.This study can solve the problems of slow growth and low metabolite synthesis of D.salina to a certain extent,and provide a reference for the cultivation of high-yield β-carotene algal strains.
