摘要 目的:从健康婴儿的肠道菌群中分离并鉴定一株短双歧杆菌,同时评估其在促进斑马鱼骨骼健康方面的益生潜力,并探讨在发酵乳制备中的潜在应用价值。方法与结果:对所分离的菌株经形态学表征、API50、16S rRNA和tuf基因比对鉴定该菌株为短双歧杆菌,命名为短双歧杆菌i1088。耐药性实验发现,i1088对万古霉素、青霉素和利福平等13种常见的抗生素较敏感。耐酸耐胆盐实验发现,该菌株在p H3.0和0.3%胆盐条件下3 h存活率均大于92%,具有较好的耐酸耐胆盐特性。在动物实验中,发现该菌株对斑马鱼幼鱼的骨骼发育具有显著促进作用(与对照组比较,P<0.05),并且还能显著改善骨质疏松模型斑马鱼的骨质状况(与模型组相比,P<0.05)。酸乳发酵实验表明,该菌株能够改善发酵乳的香气并提高整体喜好度(P<0.05)。结论:分离自健康婴儿肠道的短双歧杆菌i1088,作为一种安全且优良的益生菌,不仅在促进骨骼健康方面展现出较好的益生特性,而且在益生菌酸乳的制备上也具有广阔的应用潜力和市场前景。 Objective:The objective of this study was to isolate and identify a strain of Bifidobacterium brevis from the intestinal flora of healthy infants,evaluate its prebiotic potential for bone health,and investigate its potential application in the production of fermented milk.Methods and Results:The isolated strain was identified as Bifidobacterium brevis through morphological characterization,API50,16S rRNA,and tuf gene comparison,and was designated as Bifidobacterium brevis i1088.Drug resistance tests demonstrated that i1088 exhibited sensitivity to 13 antibiotics,including vancomycin,penicillin,and rifamycin.Acid and bile salt resistance experiments showed that the 3-hour survival rate of the strain was greater than 92%under pH 3.0 and 0.3%bile salt conditions,indicating that the strain had good acid and bile salt resistance.In animal experiments,the strain was found to significantly enhance bone development in juvenile zebrafish(compared with the control group,P<0.05),and also notably improve bone status in the osteoporosis model group(compared with the model group,P<0.05).The fermented yogurt experiment revealed that the strain enhanced the aroma of fermented milk and increased overall preference(P<0.05).Conclusion:Bifidobacterium brevis i1088 is isolated from the intestines of healthy infants,excels as a safe and exceptional probiotic.It not only demonstrates outstanding probiotic qualities that contribute to bone health,but also offers vast application potential and promising market prospects in the production of probiotic yogurt.
