摘要
目的 探究多不饱和脂肪酸(polyunsaturated fatty acid, PUFAs)是否通过调节炎症反应影响雄性小鼠的生殖功能。方法 选取12只雄性C57BL/6野生型(WT)小鼠作为对照组,其中6只饲喂普通饲料为WT+ND组,6只饲喂高脂饲料为WT+HFD组;选取12只雄性Δ15 Des转基因(TG)小鼠作为实验组,其中6只饲喂普通饲料为TG+ND组,6只饲喂高脂饲料为TG+HFD组,饲养12周后取4组小鼠睾丸及附睾组织,采用气相色谱检测睾丸组织中脂肪酸的构成及含量,采用计算机辅助精子分析系统(computer aided sperm analysis, CASA)检测附睾组织中精子运动活力[精子总活力、直线运动精子活力、精子平均路径速度(average path velocity, VAP)、精子曲线速度(curvilinear velocity, VCL)及精子直线速度(straight-line velocity, VSL)],采用实时荧光定量PCR(real-time quantitative reverse transcription PCR,RT-qPCR)检测睾丸组织中炎症因子白细胞介素(interleukin, IL;包括IL-1β、IL-6、IL-10及IL-15)转录水平的变化。结果 高脂饲料喂养的两种品系小鼠体质量均高于普通饲料喂养的同品系小鼠(P<0.05);相比同条件喂养的WT对照组小鼠,TG实验组小鼠睾丸组织中n-6 PUFAs减少、n-3 PUFAs增多(P<0.05);TG+ND组小鼠VAP和VSL高于WT+ND组,TG+HFD组小鼠VAP和VCL高于WT+HFD组小鼠(P<0.05);与同等喂养条件下的WT对照组小鼠相比,TG实验组小鼠精子细胞数量较多,排列紧密,而近管腔内有较大量的精子聚集;TG实验组小鼠睾丸中的IL-1β、IL-6、IL-10和IL-15的转录水平低于同条件下WT对照组小鼠(P<0.05);高脂喂养会使WT对照组小鼠睾丸中炎症因子IL-6的mRNA表达上调(P<0.05),TG实验组小鼠睾丸内IL-1β、IL-6、IL-10及IL-15的mRNA表达上调(P<0.05)。结论 在慢性炎症机体内,n-6 PUFAs促使炎症水平上升、降低雄性小鼠生殖能力,而n-3 PUFAs抑制炎症水平上升、提高雄性小鼠生殖能力。
Objective To explore whether polyunsaturated fatty acids(PUFAs)can affect the reproductive function of male mice through regulating inflammatory response,using a mouse inflammation model established by feeding high-fat diet.Methods Twelve male C57BL/6 mice(WT)were selected as the control group,with 6 of them fed conventional diet(WT+ND group)and the other 6 mice fed high-fat diet(WT+HFD group).Twelve maleΔ15 Des transgenic mice(TG)were selected as the experimental group,with 6 of them fed ordinary diet(TG+ND group)and the other 6 mice fed high-fat diet(TG+HFD group).After 12 weeks of feeding,the testis and epididymis tissues of the mice were collected,and the composition and content of fatty acids in testicle tissues were detected with gas chromatography.Sperm morphology and motility in epididymal tissues,including total sperm motility,linear sperm motility,average path velocity(VAP),curvilinear velocity(VCL),and linear velocity(VSL),were detected by sperm quality analyzer(CASA).Real-time quantitative reverse transcription PCR(RT-qPCR)was used to detect changes in the transcription levels of the inflammatory factor interleukin(IL-1β,IL-6,IL-10,IL-15)in testicular tissues.Results Compared with the WT+ND and TG+ND groups,the body mass values of mice in the WT+HFD and TG+HFD groups were increased(P<0.05).Compared with the WT+ND and WT+HFD groups,n-6 PUFAs were decreased but n-3 PUFAs were increased in the TG+ND and TG+HFD groups.Compared the WT+ND group,the TG+ND group showed elevated VAP and VSL levels(P<0.05),and the TG+HFD group had elevated VAP and VCL levels than the WT+HFD group(P<0.05).Compared with the TG+HFD group,the number of sperm cells was smaller in the WT+HFD group,and the arrangement was sparse,and there was only a small amount of sperm aggregation in the proximal lumen.The transcription levels of IL-1β,IL-10 and IL-15 in testis of the TG mice(TG+ND,TG+HFD)were lower than those of the WT mice(WT+ND,WT+HFD)(P<0.05).The mRNA expression of the inflammatory factor IL-6 in the testis was up-regulated in the WT+HFD group compared to the WT+ND group(P<0.05).The mRNA expression of IL-1β,IL-6,IL-10,and IL-15 was up-regulated in the TG+HFD group compared to the TG+ND group(P<0.05).Conclusion In the body with chronic inflammation,n-6 PUFAs may increase the inflammation levels,resulting in a decrease in the fertility of male mice.Conversely,n-3 PUFAs may inhibit the increase of inflammation levels,and thus improve the fertility of male mice.
