唐古碎米荠多糖提取工艺优化及其抗氧化活性

Extraction Process Optimization and Antioxidant Activity of Polysaccharides from Cardamine tangutorum O.E.Schulz

摘要 为进行唐古碎米荠质量评价及产品开发,优化唐古碎米荠不同部位多糖的提取工艺,比较其不同部位多糖的体外抗氧化活性。该研究利用Box-Behnken试验设计优化唐古碎米荠多糖提取工艺;采用紫外-可见分光光度法测定多糖含量;通过测定多糖羟基自由基、DPPH自由基、ABTS+自由基清除能力及总还原力评价其抗氧化活性。结果表明,唐古碎米荠地上部位多糖最佳提取工艺为料液比1∶70(g/mL)、提取时间200 min、提取温度100℃,此条件下多糖得率可达11.67%;地下部位多糖最佳提取工艺为料液比1∶70(g/mL)、提取时间180 min、提取温度100℃,此条件下多糖得率可达18.71%;抗氧化活性测定结果显示,地上部位多糖的羟基自由基、DPPH自由基、ABTS+自由基的半抑制浓度(half inhibitory concentration,IC50)分别为2.15、0.49、1.47 mg/mL,地下部位多糖的羟基自由基和ABTS+自由基的IC50分别为3.47、2.68 mg/mL,不同部位多糖表现出较低的总还原力。综上,唐古碎米荠多糖具有较好的抗氧化活性,其中地上部位多糖的抗氧化能力强于地下部位多糖。 To evaluate the quality of Cardamine tangutorum O.E.Schulz and develop products,this study optimized the extraction process of polysaccharides from different parts of the plant and compared in vitro antioxidant activities of the polysaccharides.The extraction process of polysaccharides from Cardamine tangutorum O.E.Schulz was optimized by BoxBehnken experimental design.The polysaccharide content was measured by ultraviolet(UV)visible spectrophotometry.The antioxidant properties of the polysaccharides were measured by assessing the scavenging capacity of hydroxyl radicals,1,1-diphenyl-2-picrylhydrazyl(DPPH)radicals,and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic)acid(ABTS)cationic radicals of the polysaccharides and the total reducing power.The results showed that the optimal extraction process for polysaccharides from the aerial parts of Cardamine tangutorum O.E.Schulz was as follows:a materialliquid ratio of 1∶70(g/mL),extraction time of 200 min,and extraction temperature at 100℃,yielding an extraction rate of up to 11.67%.For the subterranean parts,the optimal extraction process was a materialliquid ratio of 1∶70(g/mL),extraction time of 180 min,and extraction temperature at 100℃,yielding an extraction rate of up to 18.71%.The results of the antioxidant assay showed that the half inhibitory concentrations(IC50)of hydroxyl radicals,DPPH radicals,and ABTS+radicals of polysaccharides from the aerial parts of Cardamine tangutorum O.E.Schulz were 2.15,0.49 mg/mL,and 1.47 mg/mL,respectively,and the IC50 values of hydroxyl radicals and ABTS+radicals of polysaccharides from the subterranean parts were 3.47 mg/mL and 2.68 mg/mL,respectively.Polysaccharides from different parts showed lower reducing power.In conclusion,the polysaccharides from Carda-mine tangutorum O.E.Schulz exhibited significant antioxidant activities,with those extracted from the aerial parts demonstrating stronger antioxidant capacity compared to those from the subterranean parts.
