基于TGF-β1介导SK1/S1P3信号轴探讨针刀疗法对颈椎病兔颈后肌纤维化的影响

摘要 目的:基于TGF-β1介导的SK1/S1P3信号轴,观察针刀疗法对颈椎病兔颈后肌纤维化的影响,探讨针刀疗法治疗颈椎病的可能分子机制。方法:选取兔龄6个月的9只健康雄性新西兰兔,体质量(2.0±0.5)kg,根据随机数字法分为空白组、模型组和针刀组,每组3只。空白组不进行任何造模或干预;模型组、针刀组行低头固定法造模,共进行6周。造模成功后,模型组不采取任何干预措施;针刀组采用针刀疗法选取颈后肌病灶点进行松解干预,每周1次,共进行4次干预。干预结束后1周,采用Masson染色观察颈后肌组织纤维化情况,IF法检测颈后肌相关COL1A1、COL3A1、Myf5、N-Cadherin表达量,Real-time PCR、Western blot法检测颈后肌相关TGF-β1、SMAD2、S1P3、Rho-c mRNA和蛋白表达水平。结果:Masson染色结果显示,与空白组相比,模型组肌纤维破坏明显;与模型组相比,针刀组肌纤维化相对减轻。IF检测结果显示,与空白组比较,模型组COL1A1、COL3A1、Myf5、N-Cadherin等平均光密度明显减弱;与模型组比较,针刀组COL1A1、COL3A1、Myf5、N-Cadherin等平均光密度明显增强。Real-time PCR检测结果显示,与空白组比较,模型组TGF-β1、SMAD2、S1P3、Rho-c等mRNA表达量显著升高;与模型组比较,针刀组TGF-β1、SMAD2、S1P3、Rho-c等mRNA表达量显著降低(P<0.05)。Western blot结果显示,与空白组比较,模型组TGF-β1、SMAD2、S1P3、Rho-c等蛋白表达显著上调;与模型组比较,针刀组TGF-β1、SMAD2、S1P3、Rho-c等蛋白表达显著下调。结论:针刀疗法松解颈后肌病灶点能通过调节TGF-β1介导SK1/S1P3信号轴,改善颈后肌纤维化程度,发挥对颈椎病的治疗作用。 Objective:Based on TGF-β1-mediated SK1/S1P3 signal axis,to observe the effect of acupotomy on posterior cervical muscle fibrosis in rabbits with cervical spondylosis,so as to explore the possible molecular mechanism of acupotomy in the treatment of cervical spondylosis.Methods:9 healthy male New Zealand rabbits aged 6 months and weighing(2.0±0.5)kg were selected and divided into blank group,model group and acupotomy group according to random number method,with 3 rabbits in each group.No modeling or intervention was performed in the blank group.The model group and needle knife group were made by low head fixation method for a total of 6 weeks.After successful modeling,the model group did not take any intervention measures.In the acupotomy group,acupotomy therapy was used to select the focal points of cervical posterior myopathy for release intervention,once a week,and a total of 4 interventions were performed.1 week after the intervention,Masson staining was used to observe the fibrosis of the posterior cervical muscle tissue,and the expression levels of COL1A1,COL3A1,Myf5 and N-Cadherin related to the posterior cervical muscle were detected by IF method.The mRNA and protein expression levels of TGF-β1,SMAD2,S1P3 and Rho-c were detected by Real-time PCR and Western blot.Results:Masson staining showed that the damage of muscle fibers was obvious in the model group compared with the blank group.Compared with the model group,the muscle fibrosis of acupotomy group was relatively reduced.IF detection results showed that the average optical density of COL1A1,COL3A1,Myf5,N-Cadherin and other proteins in the model group decreased significantly compared with the blank group.Compared with the model group,the average optical density of COL1A1,COL3A1,Myf5 and N-Cadherin proteins in the acupotomy group was significantly increased.Real-time PCR results showed that the mRNA expressions of TGF-β1,SMAD2,S1P3 and Rho-c in the model group were significantly increased compared with the blank group.Compared with model group,mRNA expressions of TGF-β1,SMAD2,S1P3 and Rho-c in acupotomy group were significantly decreased(P<0.05).Western blot results showed that compared with the blank group,the expressions of TGF-β1,SMAD2,S1P3 and Rho-c in the model group were significantly up-regulated.Compared with model group,the expressions of TGF-β1,SMAD2,S1P3 and Rho-c in acupotomy group were significantly down-regulated.Conclusion:Acupotomy can relieve the focal point of posterior cervical myopathy by regulating TGF-β1-mediated SK1/S1P3 signal axis,improve the degree of fibrosis of posterior cervical muscle,and play a role in the treatment of cervical spondylosis.
