铁甲草多糖的分离纯化结构表征及其抑制HepG2细胞增殖的研究

摘要 以铁甲草为原料,采用热水浸提法提取铁甲草粗多糖(CMLCP)并用DEAE-52型纤维柱对其进行分离纯化得CMLCP-A、CMLCP-B和CMLCP-C组分;进一步测定4种组分的总糖、蛋白质和糖醛酸的含量,采用紫外分光光度计、傅立叶红外光谱和扫描电镜鉴定其结构,采用MTT法测定CMLCP对HepG2细胞存活率的影响。结果表明,CMLCP、CMLCP-A、CMLCP-B和CMLCP-C这4种组分的总糖质量分数分别为83.33%,29.43%,27.34%和26.93%,糖醛酸质量分数分别为58.00%,8.40%,8.34%和8.56%,蛋白质质量分数分别为15.21%,0.20%,0.20%和0.19%。进一步对4种组分进行结构鉴定发现,4种组分中只有CMLCP含少量核酸和蛋白质,且4种组分都具有多糖-OH和-CH的伸缩振动特征吸收峰。体外细胞试验结果表明,CMLCP对HepG2细胞增殖有明显的抑制作用且呈质量浓度依赖性,当CMLCP质量浓度为800μg/mL时,细胞存活率降低至47%,表明CMLCP能够抑制HepG2细胞的增殖。 Using Cassia mimosoides Linn.as raw material,crude polysaccharide from Cassia mimosoides Linn.(CMLCP)was extracted using hot water extraction,and CMLCP-A,CMLCP-B and CMLCP-C components were separated and purified by DEAE-52 fiber column.The contents of total sugar,protein and uronic acid of the four components were determined and their structures were identified by Ultraviolet spectrophotometer,Fourier infrared spectroscopy and scanning electron microscope.The effect of CMLCP on the survival rate of HepG2 cells was determined by MTT assay.The results showed that the total sugar content of CMLCP,CMLCPA,CMLCP-B and CMLCP-C was 83.33%,29.43%,27.34%and 26.93%,respectively,the uronic acid content was 58.00%,8.40%,8.34%and 8.56%,respectively,and the protein content was 15.21%,0.20%,0.20%and 0.19%,respectively.Further structural identification of the four components showed that only CMLCP contained a small amount of nucleic acid and protein,and all the four components had the stretching vibration characteristic absorption peaks of polysaccharide-OH and-CH.In vitro cell experiments showed that CMLCP significantly inhibited the proliferation of HepG2 cells in a concentration-dependent manner.When the concentration of CMLCP was 800μg/mL,the cell survival rate decreased to 47%,indicating that CMLCP could inhibit the proliferation of HepG2 cells.
