无果枸杞芽茶提取物与唑类药物联用对光滑念珠菌的抗菌作用

Antibacterial effect of the fruitless wolfberry sprout tea extract combined with azoles against smooth candida glabrata

摘要 目的探讨无果枸杞芽茶提取物(FWE)与唑类药物联用对光滑念珠菌的体外抗菌作用。方法采用微量液体稀释法测定3种唑类药物(氟康唑、伊曲康唑、伏立康唑)及无果枸杞芽茶提取物对52株光滑念珠菌的最小抑菌浓度(MIC);用棋盘稀释法检测无果枸杞芽茶提取物与3种唑类药物联用对8株光滑念珠菌的体外抗菌作用,并通过分级抑制浓度指数(FICI)对联合用药的效果进行评价。结果MIC显示所有菌株对氟康唑为剂量依赖敏感(MIC<32μg/mL),3株菌(5.77%)对伊曲康唑为非野生型(MIC≥4μg/mL),10株菌(19.23%)对伏立康唑为非野生型(MIC≥0.25μg/mL),其中3株菌同时对伊曲康唑和伏立康唑为非野生型。联合药敏实验表明,FWE与氟康唑联用对1株菌表现为协同作用,与伊曲康唑和伏立康唑联用分别对3株和5株菌表现为协同作用。联合用药可以将3种唑类药物对光滑念珠菌的MIC降低1/8~1/2。结论FWE与3种唑类药物联用可以协同有效地降低唑类药物的用药剂量,提示FWE可以作为一种有效的抗真菌增效剂。 Objective To investigate the antibacterial effect in vitro of fruitless wolfberry sprout tea extract(FWE)in combination with azole against smooth Candida.Methods The minimum inhibitory concentration(MIC)of three azoles(fluconazole,itraconazole,and voriconazole)as well as FWE alone against 52 isolates of Candida smoothie was determined by the micro liquid dilution method.The in vitro antimicrobial effect of FWE in combination with three azoles against eight isolates of Candida smoothie was examined by checkerboard dilution method and the combined effect was evaluated by graded inhibitory concentration index(FICI).Results The MIC values showed that all of them were dose-dependently sensitive to fluconazole(MIC<32μg/mL),three strains(5.77%)were non-wild-type to itraconazole(MIC≥4μg/mL),and 10 strains(19.23%)were non-wild-type to voriconazole(MIC≥0.25μg/mL),among which,three strains were non-wild-type to itraconazole and voriconazole at the same time.Combined drug sensitization experiments showed that the FWE showed synergistic effects with fluconazole on one strain,and with itraconazole and voriconazole on three and five strains,respectively.The combination reduced the MIC values of the three azoles against Candida glabrata by 1/8~1/2.Conclusion Combination of FWE and three azoles can synergistically and effectively reduce the dosage of azoles,suggesting that fruitless wolfberry bud tea extract can be used as an effective antifungal synergist.
