基于网络药理学探究枸杞有效成分治疗骨质疏松症的作用机制

摘要 基于网络药理学探讨枸杞有效化学成分治疗骨质疏松症的作用机制。通过TCMSP、Drug Bank、Gene Cards等数据库筛选枸杞活性成分及其潜在靶点,并结合骨质疏松症相关靶点进行交集分析,构建药物成分—靶点—基因互作网络和蛋白质-蛋白质互作网络。利用GO功能注释和KEGG通路富集分析,解析枸杞活性成分的潜在生物学功能和信号通路。筛选出枸杞活性成分44个,与骨质疏松症相关的交集靶点86个。GO分析显示,这些靶点显著富集于“营养水平响应”“氧化应激响应”等生物过程;KEGG分析表明,枸杞可能通过调控TNF信号通路、IL-17信号通路及AGE-RAGE信号通路等多条关键通路,对骨质疏松症产生抗炎、抗氧化及免疫调节作用。本研究从网络药理学角度揭示了枸杞在骨质疏松症预防治疗中的潜在作用机制,为开发基于枸杞的天然药物提供了参考。 This study aims to explore the mechanisms of active chemical components of Lycium chinense Miller in the treatment of osteoporosis using a network pharmacology approach.Active components of Lycium chinense Miller and their potential targets were identified through databases such as TCMSP,DrugBank,and GeneCards.Osteoporosis-related targets were collected and intersected with the drug targets to construct a compound-target-gene interaction network and a proteinprotein interaction(PPI)network.Gene Ontology(GO)enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis were performed to elucidate the biological functions and signaling pathways associated with the active components.A total of 44 active components of Lycium chinense Miller and 86 intersecting targets related to osteoporosis were identified.GO analysis revealed that these targets were significantly enriched in biological processes such as“response to nutrient levels”and“response to oxidative stress.”KEGG pathway analysis indicated that Lycium chinense Miller may exert anti-inflammatory,antioxidant,and immunomodulatory effects on osteoporosis through key pathways,including the TNF signaling pathway,IL-17 signaling pathway,and AGE-RAGE signaling pathway.This study elucidates the potential mechanisms by which Lycium chinense Miller exerts preventive and therapeutic effects on osteoporosis from a network pharmacology perspective,providing a reference for the development of Lycium chinense Miller-based natural medicines.
