摘要
NAC转录因子在植物抵御逆境胁迫和生长发育中扮演重要角色,GmNAC46负调控大豆对盐胁迫的应答。为了分析GmNAC46转录因子的作用机理,通过生物信息学方法、烟草瞬时表达体系和酵母双杂交方法分析GmNAC46转录因子的亚细胞定位及GmNAC46能否自身互作形成同源二聚体。生物信息学预测结果表明,GmNAC46定位于细胞核中且存在同源二聚体结构,构建了GmNAC46-pEGAD载体。瞬时表达结果表明GmNAC46定位于烟草表皮的细胞核中,说明GmNAC46是一个核定位的转录因子。构建GmNAC46-N-pGBKT7以及GmNAC46-pGADT7载体,酵母双杂交结果进一步表明GmNAC46蛋白能够以同源二聚体的形式在细胞中发挥作用。此研究结果为进一步探讨大豆GmNAC46转录因子的生物学功能和作用机理提供了理论依据。
NAC transcription factors play an important role in plant defending against stress and growth and development.GmNAC46 negatively regulates soybean response to salt stress.In order to analyze the mechanism of action of GmNAC46 transcription factors,the subcellular localization of GmNAC46 transcription factors and whether GmNAC46 can interact with each other to form homodimers were analyzed using bioinformatics methods,tobacco transient expression systems,and yeast two hybrid methods.The bioinformatics prediction results indicate that GmNAC46 is localized in the nucleus and has a homodimeric structure,and a GmNAC46-pEGAD vector was constructed.The transient expression results indicate that GmNAC46 is localized in the nucleus of tobacco epidermal cells,suggesting that GmNAC46 is a nuclear localized transcription factor.We construct GmNAC46-N-pGBKT7 and GmNAC46-pGADT7 vectors,yeast two hybrid results further indicate that GmNAC46 protein can function in cells in the form of homodimers.This research provides a theoretical basis for further exploring the biological function and mechanism of action of soybean GmNAC46 transcription factor.
