摘要
目的磁微粒化学发光产品的稳定性是磁微粒试剂的一个重要的性能,多数项目在37℃时表现不稳定。该文以自身免疫项目抗SS-A/Ro抗体免疫球蛋白G(IgG)检测试剂盒(磁微粒化学发光法)为例,探讨改善磁微粒试剂稳定性的方法。方法通过调整缓冲液的pH、使用不同蛋白封闭剂以及增加缓冲液中的盐离子浓度来探究改善该试剂稳定性的方法。结果增加缓冲液中的盐离子浓度可有效降低非特异性吸附,改善该试剂的稳定性。结论磁微粒试剂中的包被抗原有很多种,受提取方法的影响,抗原活性会存在差异。选择合适的包被抗原后,需要对所使用的包被抗原进行理化性质和提取方法以及抗原本身特性进行分析。进而能更好地选择磁微粒包被方法,提高试剂的稳定性,降低非特异性吸附,改善试剂的使用性能。
【Objective】The stability of magnetic particle chemiluminescence products is an important property of magnetic particle reagents,and most items are unstable at 37℃.In this paper,the method of improving the stability of magnetic particle reagent was discussed by taking the anti-SS-A/Ro antibody IgG detection kit(magnetic particle chemiluminescence method)as an example.【Methods】The stability of the reagent was improved by adjusting the pH of the buffer,using different protein sealers and increasing the concentration of salt ions in the buffer.【Results】Increasing the concentration of salt ions in the buffer can effectively reduce the non-specific adsorption and improve the stability of the reagent.【Conclusion】There are many kinds of coated antigens in magnetic particle reagents,and the activity of antigens varies depending on the extraction method.After selecting the appropriate coated antigen,it is necessary to analyze the physicochemical properties and extraction methods of the coated antigen used and the characteristics of the antigen itself.Thus,the magnetic particle coating method can be better selected to improve the stability of the reagent,reduce the non-specific adsorption,and improve the performance of the reagent.
