摘要
对纤维素酶提取石榴皮多糖的最佳工艺进行研究,在单因素的基础上采用正交试验研究各因素的交互影响,以多糖得率为检测指标,分别考察了料液比、pH、纤维素酶用量、酶解温度、酶解时间对石榴皮中多糖提取率的影响。实验结果表明石榴皮多糖得率影响次序为:酶解pH>酶用量>料液比>提取时间,当料液比为1∶20,酶用量为150 U/g,pH为7,时间为1 h时多糖得率达到最大,为3.13%,此条件为石榴皮多糖的最佳提取工艺。
Cellulase-degraded extraction of polysaccharides from pomegranate rind single-factor experiments were conducted to investigate the effects of five factors,such as solid-liquid ratio,cellulase concentration,pH,extraction temperature,and extraction time,on the extraction yield of polysaccharides from pomegranate rind.Using the phenol sulfuric acid method for polysaccharide content determination and taking the polysaccharide extraction rate as the index,the optimal extraction conditions were determined through orthogonal test analysis.The results showed that the order of influence of the factors was as follows:pH>cellulase dosage>solid-liquid ratio>extraction time.The optimal extraction conditions were as follows:solid-liquid ratio was 1:20,pH was 7.0,a cellulase dosage was 150 U/g,and extraction time was 60 min.Under these conditions,the polysaccharide extraction rate could reach 3.13%.
