摘要
为建立木薯华南9号组培快繁技术体系,以鲜食木薯华南9号顶芽和带腋芽茎段作为外植体,对MS基本培养基进行改良,设计了自创基本培养基ZCM,研究培养基中添加不同浓度的6-BA和IBA对增殖、生根的影响,并探讨炼苗方式对移栽成活率的作用。结果表明:外植体在MS和ZCM培养基上培养均可诱导萌芽和生长,茎段和顶芽无菌苗诱导率分别为66.7%和46.7%,外植体采用带腋芽茎段为佳。继代增殖培养的适宜培养基是MS和ZCM,增殖系数分别为3.5和3.4。适宜的生根培养基是MS和ZCM,生根率达到100%,根数多且长。组培生根苗采用先出瓶后液培炼苗方法,可有效提高移栽成活率。
In order to establish the tissue culture and rapid propagation technical system of cassava Huanan 9,the apical buds and stem segments with axillary buds of fresh cassava Huanan 9 were used as the explants to improve the MS basic medium,and the self-created basic medium ZCM was designed.Then,the effects of adding different concentrations of 6-BA and IBA to the medium on the proliferation and rooting were studied,and the effects of hardening seedling methods on the survival rate of transplanting were also discussed.The results showed that:the explants cultured on MS and ZCM medium could induce the germination and growth,and the induction rates of aseptic seedlings from stem segments and apical buds were 66.7%and 46.7%,respectively.It was better to use the stem segments with axillary buds as the explants.The suitable medium for proliferation culture was MS and ZCM,and the proliferation coefficients were 3.5 and 3.4,respectively.The suitable rooting culture medium was MS and ZCM,with the rooting rate reaching 100%,and the number of roots was large and long.Therefore,the survival rate of transplanting could be effectively improved by using the hardening seedling method of first out of the bottle and then liquid culture in the tissue culture of root seedlings.
