摘要
目的观察三仙安胃方对小鼠单次给药的毒性。方法以最大浓度和最大灌胃容积的三仙安胃方浓缩液于24 h内给药3次,观察小鼠的中毒反应和死亡情况,记录小鼠的外观、进食、排泄、活动情况和体质量,14d后取小鼠内脏组织计算脏器系数,行HE染色,进行组织病理学检查。结果预试验和最大给药实验中,给药组和对照组小鼠均未出现死亡及其他异常情况,给药前后小鼠各项观察指标差异均无统计学意义(P>0.05),HE染色无明显病理改变。结论小鼠每日的最大给药量86 g生药/kg,相当于临床人日用剂量的72倍,表明在规定剂量下服用是安全的。
Objective To observe the toxicity of Sanxian Anwei Formula(三仙安胃方,SXAWF)in mice.Methods The concentrated SXAWF was administered three times within 24 hours at the maximum concentration and maximum gavage volume.The toxic reactions and mortality of mice were observed.The appearance,feeding,excretion,activity,and body weight of mice were recorded.After 14 days,visceral tissues of mice were collected to calculate organ coefficients,and hematoxylin-eosin(HE)staining was performed for histopathological examination.Results In both the preliminary and maximum administration experiments,no deaths or abnormalities were observed in the treatment group and the blank group.There was no statistically significant differences in observation indicators of the mice before and after administration(P>0.05),and HE staining revealed no significant pathological changes.Conclusion The maximum daily dose administered to mice was 86 g of raw herb/kg,72 times the clinical daily dose for humans,indicating that the formula is safe when taken at the prescribed dosage.
