摘要
目的探究肾阳虚抑郁大鼠腓肠肌重量减低的病理机制,并阐明温阳填精解郁方肌肉量减少的作用途径。方法将40只雄性SD大鼠随机分为正常组、模型组、氟西汀组、温阳填精解郁方高剂量组、温阳填精解郁方中剂量组。正常组以外4组建立肾阳虚抑郁大鼠模型,造模成功后给予对应药物治疗。在治疗前后对各组大鼠进行行为学评价,并对大鼠腓肠肌的病理改变、PGC-1α蛋白表达、IL-6、IL-17A、TNF-α、IL-1β、IFN-γ、IL-4、IL-10、TGF-β1、MDA水平及SOD活力进行测定。结果肾阳虚抑郁大鼠呈现出懒惰的行为倾向和体重增势减缓,腓肠肌重量低于正常大鼠且肌纤维排列紊乱、变细伴炎性浸润。进一步检测发现肾阳虚抑郁大鼠腓肠肌PGC-1α水平降低,并处于低度炎症及氧化应激状态。温阳填精解郁方可以增加肾阳虚抑郁大鼠的腓肠肌重量,改善腓肠肌纤维排列紊乱、炎症浸润的病理变化,提升PGC-1α表达,缓解腓肠肌低度炎症及氧化应激。结论肾阳虚抑郁大鼠腓肠肌炎症及氧化应激可能是诱发肌肉减少的潜在病理基础。温阳填精解郁方可以上调腓肠肌PGC-1α表达,缓解低度炎症,抑制氧化应激,最终增加腓肠肌重量。
Objective To explore the pathological mechanisms underlying reduced gastrocnemius muscle(GM)weight in rats with kid⁃ney yang deficiency depression,and to elucidate the therapeutic pathways by which Wenyang Tianjing Jieyu Decoction(温阳填精解郁方,WTJD)restores muscle weight.Methods 40 male SD rats were randomly divided into the normal group,model group,fluoxetine group,WTJD high-dose and medium-dose groups.The rat model of kidney yang deficiency depression was established in the 4 groups except the normal group,and the corresponding drugs were given after the model was successfully established.The behavior of rats in each group was evaluated before and after treatment,and the pathological changes of GM,PGC-1αprotein expression,IL-6,IL-17A,TNF-α,IL-1β,IFN-γ,IL-4,IL-10,TGF-β1,MDA levels and SOD activity were measured.Results The rats with kidney yang deficiency depression showed lazy behavior tendency and the weight gain slowed down,the weight of GM was lower than that of normal rats,and the muscle fibers were arranged disorderly and thin with inflammatory infiltration.Further detection showed that the level of PGC-1αin GM of rats with kidney yang deficiency depression was decreased,with low inflammation and oxidative stress.WTJD could increase the weight of GM in rats with kidney yang deficiency depression,ameliorate the pathological conditions of GM fiber ar⁃rangement disorder and inflammatory infiltration,increase the expression of PGC-1α,and alleviate the low-grade inflammation and ox⁃idative stress of GM.Conclusion Gastrocnemius inflammation and oxidative stress may be the potential pathological basis of muscle re⁃duction in rats with kidney yang deficiency depression.WTJD can up-regulate the expression of PGC-1αin GM,alleviate low-grade inflammation,inhibit oxidative stress,thus increasing the weight of GM.
