新型松果壳碳量子点的合成及其用于四环素的快速检测

Preparation of novel carbon quantum dots from conifer cone shells and their application in rapid detection of tetracycline

摘要 目的以生物质废物松果壳为原料,通过一步水热法合成新型碳量子点(carbon quantum dots,CQDs),并将其应用于四环素的定量检测.方法利用高分辨透射电子显微镜(HRTEM)、傅里叶变换红外光谱仪(FTIR)、紫外-可见吸收光谱仪(UV-Vis)和荧光光谱仪对其进行表征和光学性质的研究.结果碳量子点颗粒呈圆形,平均尺寸为6.5 nm;最大激发波长约为365 nm,最佳发射波长约为440 nm;碳量子点在紫外光下(365 nm)呈现出蓝色荧光,在波长270 nm左右处有较强的紫外吸收带,可归因C=O的n-π^(*)跃迁;由于内滤效应,碳量子点的荧光在四环素的存在下被淬灭,碳量子点对四环素的检测具有较高的选择性.荧光强度比值(F/F_(0))与0.01~30μmol·L^(-1)内四环素浓度呈现良好的线性关系(R^(2)=0.996),检测限为0.29μmol·L^(-1).将其应用于自来水、河水和牛奶中,进行加标回收率实验,回收率在95.9%~105.2%内,RSD≤6.6%(n=3).结论制备的以松果壳为碳源的碳量子点,可以快速、高效地用于四环素的检测. Objective Novel carbon quantum dots(CQDs)were synthesized using conifer cone as raw material by one-step hydrothermal method and successfully applied to the detect tetracycline(TC).Methods High resolution transmission electron microscope(HRTEM),Fourier transform infrared spectrometer(FTIR),Visible absorption spectrometer(UV-Vis)and fluorescence spectrometer were used to characterize and study optical properties of CQDs.Results The CQDs displayed a spherical shape with the average size of 6.5 nm;The maximum excitation wavelength was about 365 nm and the optimum emission wavelength was about 440 nm;CQDs presented a strong blue luminescence pattern under ultraviolet light(λ=365 nm).Additionally,the CQDs had an absorption peak near 270 nm in the UV-Vis spectrum,which could be attributed to the n-π^(*)transition of C=O;Owing to the inner filter effect,the fluorescence intensity of CQDs was substantially quenched in the presence of tetracycline,realizing the detection of tetracycline with a high selectivity.Furthermore,it showed a good linear relationship between the F/F_(0) and TC concentrations between 0.01-30μmol·L^(-1)(R^(2)=0.996),and the limit of detection(LOD)was 0.29μmol·L^(-1).It was successfully applied to tap water,river water and milk with the satisfactory recovery of 95.9%-105.2%,and the standard deviation RSD of the measured value was not more than 6.6%(n=3).Conclusion The CQDs prepared using conifer cone as carbon source can be used for the detection of TC quickly and efficiently.
