黄精提取工艺优化与特征图谱建立

Optimization of Extraction Process of Polygonati Rhizoma and Establishment of Characteristic Chromatogram

摘要 目的:优化黄精的提取工艺,建立其特征图谱。方法:以提取时间、加水量、提取次数为考察因素进行单因素试验,并通过响应面试验优化黄精的提取工艺,以出膏率和多糖含量为评价指标并赋予权重,获得最佳提取工艺,同时建立黄精的高效液相特征图谱。结果:最佳提取工艺为提取4次、提取130 min、加9倍量水,验证实验黄精的出膏率和多糖质量分数分别为68.45%和10.28%,综合评分为101.69,与模型预测值相符。高效液相特征图谱中共出现9个共有峰,以峰4(5-羟基糠醛)为参照峰,相对保留时间的RSD均小于1,样品高效液相色谱图与特征图谱的相似度较高。结论:该提取工艺稳定可靠,指标符合要求,可为黄精的提取工艺研究提供参考,所建立的高效液相特征图谱可为黄精质量控制研究提供参考。 Objective:To optimize the extraction process of Polygonati Rhizoma and establish its characteristic spectrum.Methods:A single-factor experiment was conducted on the extraction time,amount of water added,and number of extractions.The extraction process of Polygonati Rhizoma was optimized by response surface test.The extraction rate and polysaccharide content were used as evaluation indicators,and weights were assigned to obtain the best extraction process.At the same time,a high-performance liquid chromatography(HPLC)characteristic spectrum was established for Polygonati Rhizoma.Results:The optimal extraction process was to extract four times for 130 min and add nine times the amount of water.The extraction yield and polysaccharide content of the verification experiment were 68.45%and 10.28%,respectively.The comprehensive score was 101.69,which was consistent with the model prediction value.There were nine common peaks in the HPLC characteristic spectrum,with peak 4(5-hydroxyfurfural)as the reference peak.The relative retention time RSD values were all less than 1,and the similarity between the sample′s HPLC and the characteristic spectrum is quite high.Conclusion:The extraction process is stable and reliable,and the indicators meet the requirements.It can provide a reference for the research on extraction process of Polygonati Rhizoma.The established HPLC characteristic spectrum can provide a reference for the research on quality control of Polygonati Rhizoma.
