单种营养物质赖氨酸添食对家蚕生长发育和茧质性状的影响

Effects of Single Nutrient Lysine Feeding on the Growth and Deve-lopment and Cocoon Characters of Silkworm, Bombyx mori

摘要 赖氨酸是家蚕生长发育过程中的一种必需氨基酸,需从桑叶中摄取。对3龄期家蚕幼虫添食0.01 g/mL和0.02 g/mL赖氨酸,分析赖氨酸对家蚕生长发育和茧质性状的影响。研究结果显示,0.01 g/mL赖氨酸添食组家蚕从5龄第4天起体质量显著增加,到第7天时相比于对照组(2.03 g),平均每头蚕体质量(2.14 g)增加5.41%。0.01 g/mL赖氨酸添食组雌雄家蚕的茧层率分别提高0.68和0.76百分点。对赖氨酸降解过程关键酶α-氨基半醛合酶(α-aminoadipic semialdehyde synthase, AASS)的编码基因BmAASS进行不同组织表达水平分析,发现随着添食赖氨酸浓度升高,在5龄第3天幼虫的中肠和丝腺中BmAASS的表达量逐渐增加,而在脂肪体中的表达量逐渐降低。同时,分析添食赖氨酸对丝蛋白基因表达的影响,结果显示添食赖氨酸能够促进Fib-H和Ser-1的表达,同时抑制Fib-L和P25表达。以上结果表明,添食0.01 g/mL赖氨酸对家蚕的生长发育及茧层率等茧质性状具有促进作用。因此,认为0.01 g/mL赖氨酸可以作为单种营养物质添加剂,这为家蚕人工饲料的品质优化和改善提供了实验基础和数据参考。 Lysine is an essential amino acid in the growth and development of silkworm and needs to be taken from mulberry leaf.In this study,we added 0.01 g/mL and 0.02 g/mL lysine,respectively,to the 3rd instar larvae to investigate the effects of lysine on the growth,development,and cocoon characters of silkworm.The results revealed that in the 0.01 g/mL lysine group,there was a significant increase in body weight starting from the 4th day of the 5th instar,and on the 7th day,the average body weight of each silkworm(2.14 g)increased by 5.41%compared to that of the control group(2.03 g).Additionally,both female and male silkworms in the 0.01 g/mL lysine group exhibited an increase in cocoon layer rate by 0.68 and 0.76 percentage points,respectively.We also analyzed the expression levels of the encoding gene of BmAASS(Bombyx moriα-aminoadipic semialdehyde synthase,a key enzyme involved in lysine degradation process)in different tissues.The results showed that as lysine concentration increased,Bm-AASS expression levels gradually rose in both midgut and silk gland of the 3rd day silkworm larvae of the 5th instar while decreasing in fat body.At the same time,the analysis of the effects of lysine feeding on the expressions of silk protein genes showed that lysine feeding could promote the expression of Fib-H and Ser-1,while inhibit the expressions of Fib-L and P 25.The results indicated that 0.01 g/mL lysine feeding could promote the growth and development of silkworm and the cocoon layer.This study provides valuable experimental evidence supporting the usage of 0.01 g/mL lysine as a single nutrient additive for optimizing artificial feed quality for silkworm.
