摘要
目的探讨黄芪山药组方配伍治疗脾虚腹泻的病理生理作用机制并验证其临床效果。方法随机将SD幼鼠分为五组,即空白对照组、模型组、阳性药组、黄芪山药散低剂量组、黄芪山药散高剂量组,每组4只。生大黄水煎液灌胃建立脾虚腹泻幼鼠模型,阳性药组予蒙脱石散0.945 g/kg、双歧杆菌胶囊132.3 mg/kg灌胃,黄芪山药散低剂量组予黄芪1.89 g/kg、山药6.3 g/kg灌胃,黄芪山药散高剂量组予黄芪3.78 g/kg、山药12.6 g/kg灌胃,空白对照组、模型组给予生理盐水灌胃。使用苏木精-伊红(HE)染色法观察其组织结构、细胞类型及炎症程度。结果HE染色显示,黄芪山药散高剂量组大鼠相比于模型组、黄芪山药散低剂量组,结肠黏膜有所改善,出血减少。结论黄芪山药配伍的组方对脾虚泄泻幼鼠的治疗有效可行。
Objective To explore the pathophysiological mechanism of compatibility of Radix Astragali seu Hedysari and Rhizoma Di-oscoreae in treating spleen deficiency diarrhea,and to verify its clinical effect.Methods The young SD rats were randomly divided into five groups,namely the blank control group,the model group,the positive drug group,the low-dose Huangqi Shanyao powder group,and the high-dose Huangqi Shanyao powder group,four rats in each group.A young mouse model of diarrhea due to spleen deficiency was established by intragastric administration of raw rhubarb decoction.Montmorillonite powder(0.945 g/kg)and Bifidobacterium capsule(132.3 mg/kg)were given to the positive drug group,Astragalus membranaceus(1.89 g/kg)and Dioscorea opposita(6.3 g/kg)in the low-dose Huangqi Shanyao powder group,Astragalus membranaceus(3.78 g/kg)and Dioscorea opposita(12.6 g/kg)in the high-dose Huangqi Shanyao powder group,and normal saline was given to the blank control group and model group.HE staining was used to observe the tissue structure,cell type and inflammation degree.Results HE staining showed that compared with the model group and the low-dose Huangqi Shanyao powder group,the rats in the high-dose Huangqi Shanyao powder group had improved colonic mucosa and reduced bleeding.Conclusion The compatibility of Radix Astragali seu Hedysari and Rhizoma Dioscoreae is effective and feasible for the treatment of young rats with spleen deficiency diarrhea.
