乌鳢精子超低温冷冻保存及其效果分析

摘要 为建立乌鳢(Channa argus)精子超低温冷冻保存技术,研究了不同激活液、稀释液、稀释比例、抗冻剂、保存时间对乌鳢精子冷冻保存效果的影响。结果显示:使用0.50%KCl溶液作为激活剂,精子运动时间和寿命最长,且与其他浓度激活液有显著差异。使用Hank′s或D-Hank′s作为稀释液、1∶5~1∶7的稀释比超低温冷冻保存乌鳢精子复苏效果较好。乌鳢精子超低温冷冻3 d复苏,6%~15%甘油和15%乙二醇保存精子与鲜精激活率无显著差异;冷冻5~360 d复苏,9%~15%甘油和15%乙二醇作抗冻剂保存的精子在激活率、运动时间和寿命方面均有较好表现,但与鲜精仍有显著差异。研究结果表明,使用Hank′s或D-Hank′s作为稀释液、甘油和乙二醇作抗冻剂,并选择合适稀释比,可长期有效保存乌鳢精子。 The effects of different activation solutions,diluents,dilution ratios,antifreeze agents,and preservation times on the cryopreservation efficiency of Channa argu s sperm were studied to establish Channa argus sperm cryopreservation technology.The results showed that using a 0.50%KCl solution as an activator resulted in the maximum sperm motility time and lifespan,with significant differences compared to other concentrations of KCl solutions.Using Hank′s or D-Hank′s as diluents,with a dilution ratio of 1∶5~1∶7,had a better effect on the recovery of C.argu s sperm after cryopreservation.Sperm was subjected to cryopreservation for three days for recovery,and there was no significant difference in activation rate between fresh and preserved sperm using 6%-15%glycerol and 15%ethylene glycol.Sperm that were frozen for 5-360 days and revived,and preserved with 9%-15%glycerol and 15%ethylene glycol as antifreeze agents,showed good performance in activation rate,motility time,and lifespan,but still showed significant differences compared to fresh sperm.The research results indicate that the cryopreservation technology established through the study,which uses Hank′s or D-Hank′s as the diluent,glycerol and ethylene glycol as cryoprotectants,and selects an appropriate dilution ratio,can effectively preserve C.argus sperm for long term.
