摘要
硒是人体生长发育所必需的微量元素,对人体健康起着至关重要的作用。常见的硒形态主要有亚硒酸盐、硒酸盐、硒代胱氨酸、甲基硒代半胱氨酸、硒代蛋氨酸等。通过硒形态分析可以为膳食硒摄入指南提供依据,有助于富硒农产品种植开发,所以针对生物样品中的硒形态进行分离与检测是十分必要的。对食品中硒的主要存在形式及富硒食品的定量标准进行概述,总结近年来生物样品硒形态的提取和分离检测技术。其中常用的液液萃取方式操作繁琐、耗时较长,随后发展的微波辅助消解可以显著提高提取效率;目前应用较多的样品前处理方法是蛋白酶辅助提取法,具有反应条件温和、硒形态不易转化、绿色环保等特点。高效液相色谱-电感耦合等离子体质谱联用(HPLC-ICP-MS)是近些年来元素形态分析研究中应用最多的技术,融合高效液相色谱分离能力强和电感耦合等离子体质谱线性范围宽、检出限低等优点,适用于分析检测不同基质的痕量硒。实现在线、高效、原位分析,完善不同种类硒标准物质的研制,加强化学与医学等多学科的交叉研究等是未来硒形态分析发展的重要方向。
Selenium is an essential trace element for human growth and development and plays a crucial role in human health.The common selenium forms include selenite,selenate,selenocysteine,methylselenocysteine,selenomethionine,etc.The analysis of selenium morphology can offer a basis for dietary selenium intake guidelines and contribute to the cultivation and development of selenium-rich agricultural products.Therefore,it is necessary to separate and detect selenium morphology in biological samples.This paper presents the main forms of selenium in food and the quantitative standard of selenium-rich food,and summarizes the extraction,separation,and detection techniques of selenium form in biological samples in recent years.The commonly used liquid-liquid extraction method is complex and time-consuming,while the microwave-assisted digestion developed later can significantly enhance the extraction efficiency.Currently,the most widely used sample pretreatment method is protease-assisted extraction,which features mild reaction conditions,easy transformation of selenium form,and green environmental protection.High performance liquid chromatography-Inductively coupled plasma mass spectrometry(HPLC-ICP-MS)is the most widely applied technique in elemental morphology analysis in recent years.It combines the advantages of high performance liquid chromatography(HPLC)and the wide linear range and low detection limit of inductively coupled plasma mass spectrometry(ICP-MS),and is suitable for the analysis and detection of trace selenium in different substrates.It is significant to achieve on-line,efficient,and in-situ analysis,improve the development of different kinds of selenium reference materials,and strengthen the cross-disciplinary research of chemistry and medicine.
