高pH值胁迫对中华绒螯蟹生理与组织的影响

摘要 开展了高pH值胁迫对中华绒螯蟹生理与组织的影响试验。设置试验组和对照组,试验组采用1 mol/L NaOH溶液调制养殖用水,pH值为(9.50±0.30),对照组养殖用水为充分曝气的自来水,pH值为(7.25±0.25)。分别于pH值调整后的0 d、5 d、10 d、15 d时,检测河蟹血淋巴生理代谢相关指标的变化,同时在试验第15 d时,对河蟹肝胰腺和鳃进行显微及超微观察。结果表明,高pH值胁迫下,河蟹血细胞密度(DHC)和甘油三酯(TG)含量持续下降,超氧化物歧化酶(SOD)活性和血蓝蛋白(Hc)含量呈先升高后下降的趋势,且最终极显著低于对照组(P<0.01);葡萄糖(GLU)和乳酸(LD)含量呈先升高后下降趋势,但最终均极显著高于对照组(P<0.01);在高pH值胁迫15 d时,河蟹肝胰腺、鳃等重要器官的组织结构受损。指出,高pH值胁迫下,河蟹SOD活性、DHC及Hc含量降低,血淋巴中GLU、LD及TG的异常变化,肝胰腺、鳃等重要器官的组织结构受损,最终导致河蟹正常的生存和生长受到威胁。 The study was conducted to investigate the effects of high pH stress on the physiology and tissues of the Chinese mitten crab(Eriocheir sinensis).Experimental and control groups were established,aquaculture water of the experimental group of this study was modulated to(9.50+0.30)using 1 mol/L NaOH,and control group was running water with adequate aeration,pH was(7.25±0.25).Related indicators of physiological metabolism of hemolymph were detected on 0 d,5 d,10 d,and 15 d respectively after pH modulation,and microscopic and ultrastructural observations of hepatopancreas and gills of E.sinensis were performed on 15^(th)day.The results showed that under high pH stress,the density of hemocyte and triglyceride content continued to decline,superoxide dismutase activity and concentration of hemocyanin showed the trend of increasing first and decreasing afterwards,and were lower than control groups(P<0.01);Concentrations of glucose and lactic acid increased at first and then decreased,but were extremely higher than control groups(P<0.01).Histological structures of hepatopancreas and gills were affected under high pH stress for 15 d.The study revealed that high pH stress significantly reduced superoxide dismutase activity,the density of hemocyte and hemocyanin levels in Chinese mitten crab,while causing abnormal fluctuations in hemolymph glucose,lactate dehydrogenase and triglyceride,and the structural damage to hepatopancreas and gills,ultimately threatening normal survival and growth of E.sinensis.
