摘要
目的对云南省普洱市江城县的10个地衣来源内生真菌的多样性进行勘探,旨在丰富地衣内生真菌的多样性,并为特境微生物来源活性先导分子的发现提供菌种资源。方法本文利用内转录间隔区(ITS)序列测定和在NCBI基因库中进行BLAST比对分析,对分离出的真菌进行初步鉴定,然后对真菌的发酵产物进行抗菌和抗肿瘤细胞活性测试。结果本文从地衣中共分离得到130株内生真菌,并对它们的固体发酵提取物进行活性筛选,其中24株具有抗菌活性,5株具有显著的抗胰腺癌细胞MIA paca2活性。结论本研究进一步丰富了地衣内生真菌的种类,同时,抗菌活性及抗肿瘤细胞活性菌株的发现为挖掘特殊生境来源真菌的活性次级代谢产物提供了菌株资源。
Objective To investigate the diversity of endophytic fungi derived from 10 lichens in Jiangcheng County of Puer City,Yunnan Province,with the aim of exploring the diversity of endophytic fungi sourced from lichen and providing microbial resources for the discovery of lead compounds from the special habitat microorganisms.Methods The fungi were preliminary identified by the sequence determination of the internal transcribed spacer(ITS)and BLAST alignment analysis in the NCBI gene library,and the fermentation extracts were evaluated for the antibacterial and anticancer cell activities.Results A total of 130 strains of endophytic fungi were isolated.Among them,24 strains showed antibacterial activity,and 5 strains exhibited significant cytotoxicity with the pancreatic cancer cell of MIA paca2.Conclusion This study further enriched the species of endophytic fungi in lichens,and the discovery of strains with antibacterial activity and anticancer cell activity provided strain resources for further exploring the active secondary metabolites from the special habitat fungi.
