摘要
为研究松材线虫侵染对樟子松抗性生理特征的影响,以3年生樟子松为试验对象,在人工接种松材线虫后的1、3、7、15、30 d,测定其针叶内过氧化氢酶(CAT)、过氧化物酶(POD)和超氧化物歧化酶(SOD)活性,信号转导分子过氧化氢、水杨酸和茉莉酸含量,渗透调节物质(可溶性蛋白、可溶性糖和游离脯氨酸)含量、类黄酮含量的动态变化。结果表明:抗氧化酶中POD活性先升高后降低,接种3 d时活性处于最高水平且与对照组之间有极显著差异(P<0.01)。过氧化氢含量持续降低,在1 d时与对照组之间达到极显著差异(P<0.01)。水杨酸含量持续高于对照组,且达到极显著差异(P<0.01)。茉莉酸含量先降低,1 d、3 d与对照组达到极显著差异(P<0.01)。可溶性蛋白含量呈先升高后降低再增高的趋势,可溶性糖含量变化趋势则相反。游离脯氨酸和类黄酮含量先升高后降低,在接种第15 d与对照组达到极显著差异(P<0.01)。以上的变化显示,松材线虫的侵染诱导樟子松产生了应激反应,抗氧化系统、渗透调节系统和类黄酮发生动态变化,共同抵抗松材线虫的侵染。
To investigate the effect of Bursaphelenchus xylophilus infestation on the resistance physiological characteristics of Pinus sylvestris var.mongolica,3-year-old Pinus sylvestris var.mongolica trees were used as experimental subjects.The activities of catalase(CAT),peroxidase(POD),and superoxide dismutase(SOD)in the needles,the contents of signaling molecules hydrogen peroxide(H2O2),salicylic acid,and jasmonic acid,the contents of osmoregulatory substances(soluble proteins,soluble sugars,and free proline),and the dynamic changes in flavonoid contents were measured at 1,3,7,15,and 30 days after artificial inoculation with Bursaphelenchus xylophilus.The results showed that the activity of POD in the antioxidant enzyme increased first and then decreased,with the highest activity at 3 days after inoculation and a significant difference compared to the control group(P<0.01).The H2O2 content continued to decrease,reaching a significant difference compared to the control group at 1 day(P<0.01).The salicylic acid content continued to be higher than the control group,reaching a significant difference(P<0.01).The jasmonic acid content first decreased,reaching a significant difference compared to the control group at 1 and 3 days(P<0.01).The soluble protein content first increased,then decreased,and then increased again,while the soluble sugar content had the opposite trend.The free proline and flavonoid contents first increased and then decreased,reaching a significant difference compared to the control group at 15 days after inoculation(P<0.01).These changes indicate that the infestation of Bursaphelenchus xylophilus induced a stress response in the Pinus sylvestris var.mongolica,with dynamic changes in the antioxidant system,osmoregulatory system,and flavonoids,which worked together to resist the infestation of Bursaphelenchus xylophilus.
