不同配比的正己烷-乙醇脱脂溶剂对黄粉虫幼虫蛋白结构和功能特性的影响

Effects of different proportions of n-hexane-ethanol defatting solvents on structural and functional properties of Tenebrio molitor larvae protein

摘要 探究不同体积配比正己烷-乙醇(正己烷:乙醇为10:0、8:2、6:4、4:6、2:8、0:10)的脱脂溶剂对黄粉虫幼虫蛋白结构和功能特性的影响,结果表明:当脱脂溶剂中乙醇体积占比较低时(10:0、8:2、6:4),黄粉虫幼虫蛋白脱脂率随乙醇比例增加显著升高(P<0.05),蛋白质结构展开,巯基质量摩尔浓度和表面疏水性显著增加(P<0.05),同时,黄粉虫幼虫蛋白的溶解性、持水持油性、乳化性、凝胶强度和保水性均显著增强(P<0.05)。随着脱脂溶剂中乙醇体积占比进一步增加(4:6、2:8、0:10),黄粉虫幼虫蛋白质发生变性聚集,功能特性变差。研究明确了应用于黄粉虫幼虫蛋白的正己烷-乙醇脱脂溶剂最佳体积混合配比为6:4,为黄粉虫幼虫蛋白在食品加工方面的应用提供参考。 The study investigated the effects of defatting solvents with varying volume ratios of n-hexane to ethanol(10:0,8:2,6:4,4:6,2:8,0:10)on the protein structure and functional properties of Tenebrio molitor larvae protein.Results demonstrated that when the ethanol proportion in the defatting solvent was low(10:0,8:2,6:4),the defatting efficiency increased significantly(P<0.05)with rising ethanol content.Concurrently,the protein structure unfolded,accompanied by marked increases in sulfhydryl group content and surface hydrophobicity(P<0.05).These structural changes enhanced solubility,water/oil holding capacity,emulsifying activity,gel strength,and water retention(P<0.05).However,further increases in ethanol proportion(4:6,2:8,0:10)induced protein denaturation and aggregation,leading to deteriorated functional properties.The optimal n-hexane-ethanol ratio for Tenebriomolitor larval protein defatting was determined as 6∶4,providing a reference for its application in food processing.
