南方水稻黑条矮缩病毒的检测及遗传多样性分析

摘要 为明确南方水稻黑条矮缩病在海南省发生情况及其病原--南方水稻黑条矮缩病毒(Southern rice black-streaked dwarf virus,SRBSDV)变异情况,采用RT-PCR方法对海南省5个地区表现为矮缩和叶色浓绿症状的154份水稻样品进行检测。结果表明:SRBSDV平均检出率为50.12%。对其中7份样品的SRBSDV外壳蛋白(Coatprotein,CP)基因序列进行克隆和遗传多样性分析,7条CP序列核苷酸和编码蛋白氨基酸序列一致性分别为99.51%~99.82%和99.23%~99.72%,与NCBI数据库中获取的其他15条SRBSDV分离物的CP基因核苷酸和氨基酸序列一致性分别为97.82%~99.83%和97.50%~99.21%。构建系统发育进化树发现,SRBSDVCP基因序列与来自中国(中国的江苏),(登录号:HQ916872)和越南(登录号:KM454846)序列聚为一簇,亲缘关系较近。遗传距离分析显示,7条序列与中国(中国的江西)分离物(登录号:JQ927009)的遗传距离为0.002~0.005,差异不显著。 To elucidate the occurrence of Southern rice black-streaked dwarf virus(SRBSDV)in Hainan Province and the variation of its pathogen,RT-PCR was used to detect 154 rice samples from five regions of Hainan Province that exhibiting symptoms of stunting and dark green leaves.The results indicated that the average detection rate of SRBSDV was 50.12%.The cloning and genetic diversity analysis of the SRBSDV coat protein(CP)gene sequences from seven samples showed that the identities of nucleotide sequence and amino acid sequences of the encoded protein were 99.51%-99.82%and 99.23%-99.72%,respectively.The nucleotide and amino acid sequences of CP gene were 97.82%-99.83%and 97.50%-99.21%,respectively with other 15 SRBSDV isolates obtained from the NCBI database.Phylogenetic analysis revealed that the SRBSDV CP gene sequence in this study was clustered closely with the SRBSDV CP gene sequence from Jiangsu in China(Accession number:HQ916872)and Vietnam(Accession number:KM454846),and the genetic relationship was close.Genetic distance analysis further revealed that the genetic distance between the seven sequences in this study and the Jiangxi in China isolate(Accession number:JQ927009)was 0.002-0.005,and indicating no significant differences.
