摘要
目的 探讨通督灸调节Keap1/Nrf2/ARE信号通路对急性脑缺血再灌注大鼠氧化应激损伤的影响。方法 将90只SD大鼠分为假手术组、模型组、模型+Nrf2抑制剂全反式维A酸(ATRA)组(10 mg/kg)、依达拉奉组(3 mg/kg)、通督灸组(督脉艾灸)和通督灸+ATRA组(督脉艾灸+ATRA 10 mg/kg)。构建右侧大脑中动脉栓塞(MCAO)模型,各组给予相应的干预3 d。采用Longa评分标准观察神经功能损伤情况,TTC染色法测定脑梗死体积百分比,HE染色观察脑缺血区皮层组织的病理改变,采用WST-8比色法和硫代巴比妥酸(TBA)法测定血清中超氧化物歧化酶(SOD)活性以及丙二醛(MDA)含量,2′,7′-二氯荧光素二乙酸酯(DCFH-DA)染色法分析脑组织活性氧(ROS)含量,缺血区皮层组织中通路相关Keap1、Nrf2、HO-1、NQO1蛋白表达水平使用蛋白质印迹法(Western blotting)检测。结果 通督灸组相较于模型组大鼠,脑缺血区皮层组织病理改变明显减轻,神经功能缺损评分、脑梗死体积百分比、MDA含量、Keap1蛋白表达均下调,同时在SOD活性、胞核Nrf2/Nrf2、HO-1、NQO1蛋白表达上均增加(P<0.05);在施以ATRA干预后,减弱了通督灸对于急性脑缺血再灌注大鼠的治疗作用(P<0.05)。结论 通督灸对于改善急性脑缺血再灌注大鼠大脑中的氧化应激损伤具有显著作用,这种作用可能与Keap1/Nrf2/ARE信号通路的上调有关。
Objective:To investigate the effect of Tongdu moxibustion on oxidative stress injury in acute cerebral ischemia-reperfusion(CIR)rats by regulating the Keapl/Nrf2/ARE signaling pathway.Methods:90 SD rats were divided into sham operation group,model group,model+Nrf2 inhibitor all-trans retinoic acid(ATRA)group(10 mg/kg),edaravone group(3 mg/kg),Tongdu moxibustion group,and Tongdu moxibustion+ATRA group(10 mg/kg).Neurological function was assessed using Longa scoring criteria,and brain infarct volume was measured by TTC staining.Pathological changes in the ischemic cortex were observed by HE staining.Superoxide dismutase(SOD)activity and malondialdehyde(MDA)content in serum were measured using the WST-8 colorimetric method and thiobarbituric acid(TBA)method,respectively.Reactive oxygen species(ROS)levels in brain tissue were analyzed using the 2',7'-dichlorofluorescin diacetate(DCFH-DA)staining method.Protein expression levels of Keapl,Nrf2,HO-1,and NQO1 in the ischemic cortex were detected by Western blotting.Results:Compared with the model group,the Tongdu moxibustion group showed significantly reduced pathological changes in the ischemic cortex,decreased neurological deficit scores,infarct volume percentage,MDA content,and Keapl protein expression,while increased SOD activity,nuclear Nrf2/Nrf2,HO-1,and NQO1 protein expression(0.05).After ATRA intervention,the therapeutic effects of Tongdu moxibustion on acute cerebral ischemia-reperfusion rats were weakened(P<0.05).Conclusion:Tongdu moxibustion has a significant effect on reducing oxidative stress injury in acute cerebral ischemia-reperfusion rats,which may be related to the upregulation of the Keapl/Nrf2/ARE signaling pathway。
