摘要
目的本研究旨在探讨同源框C6(HOXC6)在前列腺癌中的表达水平及其与患者临床病理特征的关系。方法运用GEPIA数据库预测HOXC6在前列腺癌组织和正常前列腺组织中的表达水平差异,并探讨了差异对患者预后的影响。收集扬州大学附属医院2021年1月至2023年9月行前列腺癌根治术治疗的83例前列腺癌患者的癌组织及对应的癌旁组织标本,通过免疫组织化学染色技术(IHC)检测了HOXC6蛋白在前列腺癌组织和癌旁组织的表达水平,并探讨HOXC6的表达水平与前列腺癌患者临床病理特征之间的相关性。结果数据库预测结果显示,HOXC6在前列腺癌组织中的表达水平较正常前列腺组织显著升高,且HOXC6高表达组的患者生存率较低于低表达组的患者(P<0.05)。免疫组化染色结果提示,HOXC6在前列腺癌细胞中主要表达于细胞质,在前列腺癌组织中,HOXC6的表达水平显著高于癌旁组织,且差异有统计学意义(P<0.001);在对不同年龄组、是否存在排尿疼痛、脉管侵犯、神经侵犯以及TNM分期的前列腺癌患者中HOXC6表达水平比较后发现,差异无统计学意义(P>0.05);不同肿瘤体积、分化程度、pT分期、淋巴结转移情况、PSA水平及Gleason分级的患者HOXC6表达比较,差异有统计学意义(P<0.05)。结论HOXC6在前列腺癌患者肿瘤组织中呈高表达,且HOXC6高表达的患者总体预后较差,提示HOXC6可作为前列腺癌诊疗的一项潜在生物标志物。
Objective To investigate the expression level of homeobox C6(HOXC6)in prostate cancer and its relationship with the clinicopathological characteristics of patients.Methods GEPIA database was used to predict the differences in the expression level of HOXC6 in prostate cancer tissues and normal prostate tissues,and the effect of the differences on the prognosis of patients was explored.The cancer tissues and corresponding paracancerous tissues of 83 patients with prostate cancer who underwent radical prostatectomy in the Affiliated Hospital of Yangzhou University from January 2021 to September 2023 were collected.The expression level of HOXC6 protein in prostate cancer tissues and paracancerous tissues was detected by immunohistochemical staining(IHC).The correlation between the expression level of HOXC6 and the clinicopathological characteristics of prostate cancer patients was investigated.Results The database prediction results showed that the expression level of HOXC6 in prostate cancer tissues was significantly higher than that in normal prostate tissues,and the survival rate of patients in the HOXC6 high expression group was lower than that in the low expression group(P<0.05).Immunohistochemical staining results suggested that HOXC6 was mainly expressed in the cytoplasm of prostate cancer cells,and the expression level of HOXC6 in prostate cancer tissues was significantly higher than that in adjacent tissues,and the difference was statistically significant(P<0.001).There was no significant difference in the expression level of HOXC6 in prostate cancer patients with different age groups,with or without urination pain,vascular invasion,nerve invasion and TNM stage(P>0.05).There were significant differences in HOXC6 expression among patients with different tumor volume,differentiation degree,pT stage,lymph node metastasis,PSA level and Gleason grade(P<0.05).Conclusion HOXC6 is highly expressed in the tumor tissues of prostate cancer patients,and the overall prognosis of patients with high expression of HOXC6 is poor,suggesting that HOXC6 can be used as a potential biomarker for the diagnosis and treatment of prostate cancer.
