“王氏止鼾方”对慢性间接缺氧小鼠肠道菌群及血清氧化三甲胺的影响

摘要 目的探讨王氏止鼾方对慢性间歇性缺氧(CIH)模型小鼠肠道菌群及体内氧化三甲胺(TMAO)含量的影响。方法C57BL/6小鼠随机分为空白对照组(Control)、模型组(CIH)、中药治疗组(CIH+TCM)和常氧治疗组(CIH+Normoxia)。除对照组外,其余各组均进行间歇性缺氧干预造模并给予相应处理。实验过程中采集小鼠肝脏、血液及粪便样本以测量各项指标。结果模型组血清TMAO含量以及肝脏FMO3蛋白和mRNA表达水平均显著高于对照组(P<0.01)。与CIH组相比,中药治疗组肝脏FMO3蛋白和mRNA表达水平明显降低(P<0.01),常氧治疗组的血清TMAO含量以及肝脏FMO3蛋白和mRNA表达也降低(P<0.01)。在肠道菌群α多样性分析中,模型组与对照组相比,Chao1、Shannon和Simpson指数差异无统计学意义(P>0.05)。而与模型组相比,中药治疗组的Shannon指数升高、Simpson指数降低(P<0.01)。主坐标分析显示各组小鼠肠道菌群的组成和结构存在显著差异。进一步的群落结构组分分析及LefSe差异性分析表明,间歇性缺氧干预及不同治疗方式后,在科、属水平上存在差异菌种。结论王氏止鼾方可调节CIH模型小鼠肠道菌群的多样性和相关菌属的丰富度平衡,这一机制可能是其防治阻塞性睡眠呼吸暂停低通气综合征(OSAHS)的重要作用途径之一。 Objective To investigate the effects of Wang's Anti-Snoring Formula on gut microbiota and serum trimethylamine N-oxide(TMAO)levels in a chronic intermittent hypoxia(CIH)mouse model.Methods C57BL/6 mice were randomly divided into four groups:control(Control),model(CI H),traditional Chinese medicine treatment(CIH+TCM),and normoxia treatment(CIH+Normoxia).Except for the Control group,all groups underwent CIH modeling and respective interventions.Liver,blood,and fecal samples were collected to measure relevant indicators.Results The CIH group showed significantly elevated serum TMAO levels and hepatic FMO3 protein/RNA expression compared to the Control group(P<0.01).The CIH+TCM group exhibited markedly reduced FMO3 protein/RNA expression versus the CIH group(P<0.01),while the CIH+Normoxia group demonstrated decreased TMAO and FMO3 levels(P<0.01).No statistical differences in Chao1,Shannon,or Simpson indices between CIH and Control(P>0.05).However,the CIH+TCM group showed increased Shannon and decreased Simpson indices(P<0.01 VS.CIH).Principal coordinate analysis showed significant differences in the composition and structure of intestinal flora among groups of mice.Further community structure component analysis and LefSe differential analysis showed that there were differences in bacterial species at the level of species and genus after intermittent hypoxia intervention and different treatment methods.Conclusion Wang's Anti-Snoring Formula may mitigate CIH-induced disturbances by modulating gut microbiota diversity and key microbial taxa,alongside suppressing TMAO generation(via FMO3 downregulation),suggesting a potential mechanism for managing obstructive sleep apnea-hypopnea syndrome(OSAHS).
