基于HPLC指纹图谱结合多元统计分析不同干燥方式下地参三萜酸类成分及其质量评价

Evaluation of Triterpenic Acids and Quality of Lycopus lucidus Under Different Drying Methods Based on HPLC Fingerprint Combined with Multivariate Statistical Analysis

摘要 目的:建立不同干燥方式下地参药材的质量评价方法。方法:采用高效液相色谱(HPLC)法建立12种不同干燥方式地参药材的指纹图谱,利用“中药色谱指纹图谱相似度评价系统”(2012版)进行相似度分析;运用层次聚类分析(HCA)、主成分分析(PCA)和正交偏最小二乘判别分析(OPLS-DA)进行化学模式识别,并测定白桦脂酸、齐墩果酸、熊果酸3种指标成分的含量。结果:12种干燥方式下地参药材HPLC指纹图谱相似度为0.866~0.974,共标定13个共有峰,指认其中3个分别为白桦脂酸、齐墩果酸和熊果酸,含量范围分别为1.268~1.818、0.107~0.250、0.416~0.802 mg/g;HCA结果显示12种干燥方式分为4类;PCA分析提取出4个主成分,累计方差贡献率达86.005%;OPLS-DA分析筛选出7个差异性标志物。结论:所建立的HPLC指纹图谱及三萜酸含量测定方法稳定可靠,可为地参药材的质量评价提供参考。 Objective:To establish a quality evaluation method for Lycopus lucidus under different drying methods.Methods:High-performance liquid chromatography(HPLC)was employed to construct the fingerprint profiles of L.lucidus samples processed by 12 different drying methods.The similarity of these fingerprints was analyzed using the“Similarity Evaluation System for Chromatographic Fingerprint of Traditional Chinese Medicine”(2012 edition).Chemical pattern recognition was performed by hierarchical clustering analysis(HCA),principal component analysis(PCA),and orthogonal partial least squares discriminant analysis(OPLS-DA).Additionally,the contents of three marker components(betulinic acid,oleanolic acid,and ursolic acid)were determined.Results:The similarity of HPLC fingerprints of L.lucidus samples processed by 12 drying methods ranged from 0.866 to 0.974.A total of 13 common peaks were identified,among which three were assigned as betulinic acid,oleanolic acid,and ursolic acid,with content ranges of 1.268 to 1.818,0.107 to 0.250,and 0.416 to 0.802 mg/g,respectively.HCA results showed that the 12 drying methods were classified into 4 categories.PCA extracted four principal components,explaining 86.005%of the cumulative variance.OPLS-DA screened out seven differential markers.Conclusion:The established HPLC fingerprint and triterpenoid acids content determination methods are stable and reliable,which can provide a reference for the quality evaluation of L.lucidus.
