毛蕊花糖苷通过TGF-β1/p38MAPK信号通路对乳腺癌细胞增殖、凋亡的影响

Influences of verbascoside on the proliferation and apoptosis of breast cancer cells through TGF-β1/p38 MAPK signaling pathway

摘要 目的研究毛蕊花糖苷对乳腺癌细胞增殖、凋亡的影响,并探讨其对转化生长因子β1(TGF-β1)/p38丝裂原活化蛋白激酶(p38MAPK)信号通路的调控作用。方法取对数生长期的MDA-MB-231细胞,分为对照组(不经任何处理),毛蕊花糖苷低(50μmol/L)、中(100μmol/L)、高(200μmol/L)浓度组,si-NC组(转染si-NC+200μmol/L毛蕊花糖苷溶液)和si-TGF-β1组(转染si-TGF-β1+200μmol/L毛蕊花糖苷溶液)。MTT法检测细胞增殖情况;流式细胞术检测细胞凋亡与细胞周期分布;Western blot法检测凋亡与细胞周期相关蛋白以及TGF-β1、p38MAPK蛋白表达;qRT-PCR法检测TGF-β1、p38MAPK基因表达。结果与对照组比较,毛蕊花糖苷各浓度组细胞活力、S期细胞比例及CyclinD1、CyclinE、Bcl-2蛋白表达量逐渐降低,细胞凋亡率,G0/G1期细胞比例、Bax蛋白表达及TGF-β1、p38MAPK基因和蛋白表达逐渐升高(P<0.05)。与毛蕊花糖苷高浓度组、si-NC组比较,si-TGF-β1组细胞活力、S期细胞比例及CyclinD1、CyclinE、Bcl-2蛋白表达量升高,细胞凋亡率、G0/G1期细胞比例、Bax蛋白表达及TGF-β1、p38MAPK基因和蛋白表达降低(P<0.05)。结论毛蕊花糖苷可能通过激活TGF-β1/p38MAPK信号通路,抑制乳腺癌细胞增殖,并促进细胞凋亡。 Objective To study the influences of verbascoside on the proliferation and apoptosis of breast cancer cells,and explore its regulatory effect on the transforming growth factorβ1(TGF-β1)/p38 mitogen-activated protein kinase(p38MAPK)signaling pathway.Methods MDA-MB-231 cells in logarithmic growth phase were taken and divided into control group(without any treatment),low(50μmol/L),medium(100μmol/L),high(200μmol/L)concentration verbascoside group,si-NC group(transfected with si-NC+200μmol/L verbascoside solution)and si-TGF-β1 group(transfected with si-TGF-β1+200μmol/L verbascoside solution).The cell proliferation was detected by MTT assay.Flow cytometry was used to detect apoptosis and cell cycle distribution.Western blot was used to detect the expressions of apoptosis and cell cycle-related proteins,and TGF-β1 and p38MAPK proteins.The expression of TGF-β1 and p38MAPK genes was detected by qRT-PCR.Results Compared with the control group,the cell viability,the proportion of cells in S phase,and the expression of CyclinDl,CyclinE and Bcl-2 protein in different concentration verbascoside groups were gradually decreased,the apoptosis rate,the proportion of cells in Go/G1 phase,the expression of Bax protein,and the gene and protein expressions of TGF-β1 and p38MAPK were gradually increased(P<0.05).Compared with the high concentration verbascoside group and si-NC group,the cell viability,the proportion of cells in S phase,and the expression of CyclinD,CyclinE and Bcl-2 protein in si-TGF-β1 group were increased,the apoptosis rate,the proportion of cells in GO/G1 phase,the expressions of Bax,and the gene and protein expressions of TGF-β1 and p38MAPK were decreased(P<0.05).Conclusion Verbasoside may inhibit the proliferation of breast cancer cells and promote cell apoptosis by activating the TGF-β1/p38MAPK signaling pathway.
