陆地棉GhCER2通过调节蜡质合成响应干旱胁迫的功能研究

摘要 ECERIFERUM 2(CER2)属于脂肪酸羟基化酶家族,参与蜡质合成过程中超长链脂肪酸(Very long chain fatty acids,VLCFAs)C_(28)的延伸过程。为揭示GhCER2调节棉花耐旱性的功能,本研究利用生物信息学、生理及分子生物学,结合扫描电镜等方法,对GhCER2的分子特征、生物学功能进行了系统分析和验证。结果发现,GhCER2在陆地棉中存在2个位于At和Dt亚基因组上的同源基因,并揭示了CER2在进化上的保守性。GhCER2启动子区包含与干旱和激素信号响应相关的顺式作用元件,且转录受干旱胁迫和ABA信号的诱导。沉默GhCER2导致棉花幼苗抗旱能力下降,抗氧化系统失衡。进一步的扫描电镜观察发现,沉默GhCER2导致棉花叶表皮蜡质晶体积累减少,叶绿素浸出和失水速率加快,降低了叶片的保水性。干旱条件下,GhCER2还通过改变气孔状态来调节叶片的蒸腾速率。本研究揭示了GhCER2通过调节叶表皮蜡质积累和气孔状态影响棉花耐旱性的调控机制。 The ECERIFERUM 2(CER2),belonging to the family of fatty acid hydroxylases,is involved in the extension process of very long chain fatty acids(VLCFAs)C_(28) during the wax synthesis process.In order to reveal the function of GhCER2 in regulating the drought tolerance of cotton,this study utilized bioinformatics,physiology and molecular biology,combined with methods such as scanning electron microscopy,to systematically analyze and verify the molecular characteristics and biological functions of GhCER2.It was found that there were two homologous genes of GhCER2 located on the At and Dt subgenomes in upland cotton,and the evolutionary conservation of CER2 was revealed.The GhCER2 promoter region contained cis-acting elements related to drought and hormonal signal responses,and its transcription was induced by drought stress and ABA signals.Silencing GhCER2 resulted in a decline in the drought resistance of cotton seedlings and an imbalance in the antioxidant system.Further electron microscopy observations of the seedlings of the GhCER2 silencing plants revealed an accumulation of waxy crystals on the epiderm leaves,an accelerated leaching and water loss rate of chlorophyll,and a decrease in the water retention of the leaves.Under drought conditions,GhCER2 also regulated the transpiration rate of leaves by changing the stomatal state.This study revealed the regulatory mechanism that GhCER2 affeded the drought tolerance of cotton by regulating the accumulation of waxy matter on the leaf epidermis and the stomatal state.
