小菜蛾抗菌肽Moricin对蝉拟青霉侵染的免疫应答

摘要 Moricin是一种重要的α-螺旋抗菌肽,在抵御病原物的侵染中发挥着重要的作用。本研究以小菜蛾Moricin基因为研究对象,通过qPCR分析,结果显示:小菜蛾Moricin基因在其整个发育阶段内均有表达,尤以4龄幼虫阶段及预蛹期的表达量较高;Moricin基因在不同组织中以脂肪体的表达量最高;当小菜蛾受到蝉拟青霉、金黄色葡萄球菌及大肠杆菌微生物的侵染时,其Moricin基因会发生显著的上调表达。沉默Moricin基因后,Moricin基因的转录水平明显下降,同时,蝉拟青霉侵染下小菜蛾幼虫的存活率也显著降低。这一结果说明Moricin在抵御病原微生物侵害中发挥着重要的作用。 Moricin is a keyα-helical antimicrobial peptide that plays a vital role in the immune defense of Plutella xylostella against pathogenic invasion.In this study,the Moricin gene of P.xylostella was selected as the target for functional analysis.Quantitative real-time PCR revealed that Moricin gene was expressed across all developmental stages,with peak expression observed in fourth-instar larvae and during the pupal stage.Among various tissues,the fat body exhibited the highest transcript levels.When the P.xylostella was infected by Isaria cicadae,Staphylococcus aureus and Escherichia coli,the expression of Moricin gene was significantly up-regulated.After silencing the Moricin gene,its transcription level markedly decreased,and the survival rate of P.xylostella larvae under I.cicadae infection was also significantly reduced.These results indicate that Moricin plays an essential role in resisting pathogenic microbial invasion.
