低含量蛋白纤维的洛蕾法鉴定

摘要 针对低含量蛋白纤维鉴别难题,探究了洛蕾法的应用。分析确定了蛋白纤维溶解条件,对比洛蕾法与常规方法的检测性能,并验证方法的重复性与检测限。试验表明:最佳溶解条件为1 mol/L NaOH溶液、70℃水浴超声15 min;洛蕾法受溶液造成质量损失、纤维显微形态难以区分、试纸显色不明显、非蛋白质氮源干扰的影响显著小于常规方法,能更有效鉴别蛋白质量分数<1%的纤维;洛蕾法测试的检测限为1.1553μg/mL。该方法具有灵敏度高、重复性好、检测快速、适用范围广等优势,为纺织产品中低含量蛋白纤维的鉴定提供了技术支撑。 The application of the Lowry method was explored for the identification of low-content protein fibers.The dissolution conditions of protein fibers were analyzed and determined,the detection performance of the Lowry method was compared with that of conventional methods,and the repeatability and detection limit of the method were verified.The experiments showed that the optimal dissolution conditions were 1 mol/L NaOH solution,ultrasonic treatment at 70°C in a water bath for 15 min.The Lowry method was significantly less affected by mass loss caused by the solution,indistinguishable fiber microscopic morphology,unclear color development of test paper,and interference from non-protein nitrogen sources than conventional methods,and could more effectively identify fibers with protein mass fraction<1%.The detection limit of the Lowry method test was 1.1553μg/mL.Lowry method has the advantages of high sensitivity,good repeatability,rapid detection,and wide applicability,providing technical support for the identification of low-level protein fibers in textile products.
