摘要
【目的】通过脂质组结合转录组分析,分析粉皮冬瓜果实上粉过程中的脂类代谢物差异和差异表达基因,为探明冬瓜果实表面蜡粉形成的物质基础和调控机制奠定基础。【方法】以粉皮冬瓜‘P131’高代自交系为材料,取授粉后10 d和20 d果实的果皮。利用UPLC-MS/MS技术,分别检测两个时期果皮中的脂类物质含量并分析差异代谢物。通过转录组测序,分析两个时期果皮中的差异表达基因,并进行KEGG和GO富集分析。【结果】从‘P131’授粉后10 d和20 d果实的果皮中共鉴定获得267种脂类物质,二者存在85种差异代谢物,其中二酰基甘油葡糖醛酸_DGDG18:3_20:3、单糖甘油二酯MGDG18:3_20:3和磷脂酰肌醇_PI16:0_18:1等在授粉后20 d的果皮中含量显著提高。转录组分析发现,授粉后10 d和20 d果实的果皮间存在343个差异表达基因,其中120个上调表达、223个下调表达,其中包括已报道与蜡粉形成相关的CER1、TPS基因等。这些差异表达基因主要富集在倍半萜和三萜生物合成途径,角质、木栓质和蜡质生物合成等与酯类物质合成相关的KEGG代谢通路,以及脂质代谢途径、超长链脂肪酸代谢途径等与脂类物质代谢相关的GO条目。【结论】初步鉴定了冬瓜果实上粉过程中的差异脂类代谢物及差异表达基因,推测冬瓜果实表面蜡粉的形成可能与酯类物质合成及脂质代谢相关。
【Objective】To explore composition and regulatory mechanism of the wax powder on the fruit surface of wax gourd,lipid metabolism and transcriptome was performed to analyze lipid and gene expression differences before and after wax loading.【Method】Wax gourd inbred line‘P131’was used as plant material and its fruit peel was cut and collected at 10 and 20 days after pollination.UPLC-MS/MS was applied to analyze the lipid composition and differences between these two stages.Transcriptome analysis was performed to understand differentially expressed genes between them.Moreover,KEGG and GO enrichment was done to understand main biological pathways involved during this period.【Result】In total,267 lipids were identified in the fruit peel of‘P131’at 10 and 20 days after pollination,and 85 lipids were found differentially accumulated between these two stages.Among these lipids,the contents of DGDG18:3_20:3,MGDG18:3_20:3 and PI16:0_18:1 were significantly higher in the fruit peel at 20 days after pollination than that at 10 days after pollination.Totally,343 genes were found differentially expressed in the fruit peel of wax gourd inbred line‘P131’at 10 and 20 days after pollination,among which 120 were up-regulated while the other 223 were down-regulated based on transcriptome analysis,and CER1,TPS and other genes previously reported involved in wax formation were included.It was found that differentially expressed genes were enriched in KEGG pathways related with ester biosynthesis,such as‘Sesquiterpenoid and triterpenoid biosynthesis’,‘Cutin,suberine and wax biosynthesis’and‘Biosynthesis of secondary metabolites’.GO enrichment analysis highlighted items related with lipid metabolism,includiung‘lipid metabolic process’and‘very long-chain fatty acid metabolic process’.【Conclusion】In conclusion,we preliminarily identified lipids and transcriptome differences during the wax loading period of wax gourd fruit.The results suggest that lipid metabolism and ester biosynthesis might relate with the process of wax powder formation.
