桑菊饮颗粒剂的制备及4种化学成分的含量测定

摘要 目的:探究桑菊饮颗粒剂的制备工艺,建立高效液相色谱色谱法(HPLC)同时测定桑菊饮颗粒剂中绿原酸、木犀草苷、芦丁和连翘苷含量的方法。方法:通过湿法制粒工艺,制备桑菊饮颗粒剂;采用HPLC法对桑菊饮颗粒中绿原酸、木犀草苷、芦丁和连翘苷的含量进行测定,色谱柱为ODS-C 18柱,流动相为乙腈-0.4%磷酸水,进行梯度洗脱,体积流量为1.0 mL/min,柱温为30℃,检测波长分别为348 nm和277 nm,进样量为5μL。结果:桑菊饮颗粒剂的制备工艺为桑菊饮浸膏∶玉米淀粉∶蔗糖=5∶10∶5,所得颗粒剂外观为浅棕色,粒径大小均匀,色泽一致,溶解性好。绿原酸、木犀草苷、芦丁和连翘苷的线性范围分别为7.03~140.67μg/mL、6.40~128.00μg/mL、6.03~120.67μg/mL和5.87~117.33μg/mL,相关系数(r)分别为0.9998、0.9998、0.9998和1;平均加样回收率分别为101.09%、100.98%、99.34%和100.34%,RSD分别为1.60%、1.87%、2.67%和2.40%。结论:桑菊饮颗粒剂制备工艺稳定可行,绿原酸、木犀草苷、芦丁和连翘苷含量测定方法准确可靠,可用于该颗粒剂的质量控制。 Objective To explore the preparation process of Sangju Yin granules and establish a high-performance liquid chromatography(HPLC)method for simultaneous determination of chlorogenic acid,luteoloside,rutin,and phillyrin content in Sangju Yin granules.Methods Prepare Sangju Yin granules through wet granulation process;HPLC method was used to determine the contents of chlorogenic acid,luteoloside,rutin,and phillyrin in Sangju Yin granules.The chromatographic column was ODS-C 18 column,and the mobile phase was acetonitrile 0.4%phosphoric acid water.Gradient elution was performed with a volume flow rate of 1.0 mL/min;The column temperature is 30℃,the detection wavelengths are 348 nm and 277 nm,and the injection volume is 5μL.Results The preparation process of Sangju Yin granules is Sangju Yin extract∶corn starch∶sucrose=5∶10∶5.The obtained granules have a light brown appearance,uniform particle size,consistent color,and good solubility.The linear ranges of chlorogenic acid,luteoloside,rutin,and phillyrin are 7.03-140.67μg/mL,6.40-128.00μg/mL,6.03-120.67μg/mL,and 5.87-117.33μg/mL,respectively,with correlation coefficients(r)of 0.9998,0.9998,0.9998,and 1,respectively;The average sample recovery rates were 101.09v%,100.98%,99.34%,and 100.34%,respectively,with RSDs of 1.60%,1.87%,2.67%,and 2.40%,respectively.Conclusions The preparation process of Sangju Yin granules is stable and feasible,and the methods for determining the contents of chlorogenic acid,luteoloside,rutin,and phillyrin are accurate and reliable,which can be used for quality control of the granules.
