基于NF-κB通路探讨负载血小板裂解物和纳米氧化铈的注射用透明质酸水凝胶对软骨细胞增殖的影响

摘要 目的探讨负载血小板裂解物和纳米氧化铈的注射用透明质酸水凝胶通过抑制核因子κB(nuclear fac-tor kappa B,NF-κB)通路活性发挥潜在的抗骨关节炎的作用。方法选取2024年3月—2024年5月东莞松山湖东华医院采购的9只SD大鼠,采用随机分组法分为3组,每组3只。以提取软骨细胞,提取的软骨细胞进行体外培养,设置对照(non-intervention,N)组、骨关节炎(osteoarthritis,OA)组、水凝胶(hyaluronic acid,HA)组。细胞培养144 h,采用实时荧光定量聚合酶链式反应法检测核因子κB抑制蛋白α(NF-kappa-B inhibitor alpha,IKB-α)、核因子-kB-p65(nuclear factor kappa B-P65,NF-κB-P65)mRNA的表达水平。结果与对照组比较,OA组的IKB-αmRNA的表达水平降低,NF-κB-P65 mRNA的表达水平升高,骨软骨细胞活性下降,差异均具有统计学意义(t=3.25,1.38,2.56;P<0.05);与OA组比较,HA组的IKB-αmRNA、NF-κB-P65表达水平及骨软骨细胞活性部分回调,差异均具有统计学意义(t=2.87,3.12,2.34;P<0.05)。结论负载血小板裂解物和纳米氧化铈的注射用透明质酸水凝胶具备潜在抗骨关节炎作用,其机制与降低NF-κB通路活性有关。 Objective To investigate the potential anti-osteoarthritis effect of hyaluronic acid hydrogel for injection loaded with platelet lysate and nano-cerium oxide by inhibiting the activity of nuclear factor kappa B(NF-κB)pathway.Methods From March 2024 to May 2024,nine SD rats purchased from Dongguan Songshan Lake Donghua Hospital were selected and randomly divided into three groups,with three rats in each group.Chondrocytes were extracted and cultured in vitro and divided into non-intervention(N)group,osteoarthritis(OA)group and hyaluronic acid(HA)group.After 144 h of cell culture,the expression levels of nuclear factor kappa B inhibitor alpha(IKB-α)and nuclear factor kappa B-p65(NF-κB-P65)mRNA were detected by real-time fluorescence quantitative polymerase chain reaction.Results Compared with the control group,the expression level of IKB-α mRNA in the OA group was decreased,the expression level of NF-κB-P65 mRNA was increased,and the activity of osteochondrocytes was decreased,with statistically significant differences(t=3.25,1.38,2.56;all P<0.05);compared with the OA group,the expression levels of IKB-α mRNA and NF-κB-P65 and the activity of osteochondrocytes in the HA group were partially reversed,and the differences were statistically significant(t=2.87,3.12,2.34;all P<0.05).Conclusion The hyaluronic acid hydrogel for injection loaded with platelet lysates and nano-ceria has potential anti-osteoarthritis effect,and its mechanism is related to the reduction of NF-κB pathway activity.
