铬胁迫对苹果组培苗生理特性的影响

摘要 为研究不同浓度的重铬酸钾对苹果砧木P1组培幼苗生理特性的影响,对其继代培养基进行4个浓度重铬酸钾25、50、100、200mg/L处理,以0mg/L为对照,测定处理后每隔7d的生理指标。结果表明,组培幼苗对25~200 mg/L铬处理均受到负面影响,负面影响与处理浓度成正相关,随处理后的时间的延长变小。幼苗体内叶绿素含量、可溶性糖含量2项指标均随铬处理浓度增大而降幅增大;同一铬浓度处理后第21 d含量开始下降,铬处理浓度越大降幅越大;抗氧化酶SOD、POD、CAT的活性,随铬处理浓度的增大而增大,随着铬处理后时间的延长均呈先升后降趋势。处理后7 d,低浓度25、50 mg/L重铬酸钾处理对幼苗影响较小,抗氧化酶活性增强幅度较小,至处理时间14d或21 d时,高浓度100、200mg/L处理的幼苗体内抗氧化酶SOD、POD、CAT活性减弱,叶绿素含量降低,幼苗受到铬胁迫毒害。 The effects of different concentrations of potassium dichromate on the physiological characteristics of P1 tissue cultured seedlings of apple rootstock were studied.The subculture medium was treated with potassium dichromate at four concentrations of25,50,100 and 200 mg/L,with O mg/L as the control.Physiological indicators were measured every 7 days after treatment.The results showed that tissue cultured seedlings were negatively affected by 25~200 mg/L chromium treatment,which was positively correlated with the treatment concentration and varied with the extension of treatment time until it decreased.The chlorophyll content in seedlings decreases with increasing treatment concentration;After the same treatment,the content began to decrease on the 21st day,and the greater the chromium treatment concentration,the greater the decrease;The activities of soluble sugars,antioxidant enzymes SOD,POD and CAT increased with the increase of chromium treatment concentration,and showed a trend of first increasing and then decreasing with the prolongation of chromium treatment time.After 7 days of treatment,low concentrations of 25 and 50 mg/L potassium dichromate had little effect on the seedlings,and the enhancement of antioxidant enzyme activity was relatively small.At 14 or 21 days of treatment,the activities of antioxidant enzymes SOD,POD and CAT in the seedlings treated with high concentrations of 100 and 200 mg/L decreased,and the chlorophyll content decreased.The seedlings were subjected to chromium stress toxicity.
