火棘含硒黄酮的离子液体超声辅助提取工艺优化及表征

摘要 含硒黄酮结合了硒和黄酮类化合物生物活性,使其在功能食品和医药领域具有广阔的应用前景。文章研究了离子液体超声辅助法提取火棘硒黄酮的工艺条件,利用响应面优化得到以总黄酮提取得率为响应值的工艺参数:提取温度50℃,超声功率150 W,提取时间42 min,料液比16 mL/g,离子液体浓度9.5 g/L,总黄酮提取得率为2.37%。纯化后总黄酮含量达51.82%,总硒含量为3.8μg/g。FTIR分析显示火棘含硒黄酮提取物中可能含有C-Se键。经HPLC-MS/MS分析,火棘含硒黄酮中主要含有高车前素、橘皮素、黄芩苷、3,4’-二羟基黄酮、羟基芜花素、地奥司明、高车前苷、木蝴蝶苷、半齿泽兰素、棕矢车菊素等黄酮类化合物。研究结果为火棘含硒黄酮的制备和进一步研究利用提供依据。 Selenium-containing flavonoids,which combine the bioactivities of selenium and flavonoids,exhibit broad application prospects in the fields of functional foods and pharmaceuticals.This study investigated the extraction conditions of selenium-containing flavonoids from Pyracantha fortuneana using an ionic liquid-based ultrasound-assisted method.The process parameters were optimized via response surface methodology(RSM)with total flavonoid extraction yield as the response value.The optimized conditions were as follows:extraction temperature of 50℃,ultrasonic power of 150W,extraction time of 42 min,solid-to-liquid ratio of 16 mL/g,and ionic liquid concentration of 9.5g/L,resulting in a total flavonoid extraction yield of 2.37%.After purification,the total flavonoid content reached 51.82%,and the total selenium content was 3.8μg/g.Fourier transform infrared spectroscopy(FTIR)analysis suggested the potential presence of C-Se bonds in the extracted selenium-containing flavonoids from Pyracantha fortuneana.According to HPLC-MS/MS analysis,the selenium-containing flavonoids from Pyracantha fortuneana mainly contain hispidulin,tangeretin,baicalin,3,4’-dihydroxyflavone,hydroxygenkwanin,diosmin,homoplantaginin,oroxin B,eupatorine,jaceosidin and other flavonoids.The findings of this study provide a basis for the preparation and further investigation and utilization of selenium-containing flavonoids from Pyracantha fortuneana.
